摘要
目的分析福建省2015年一起流行性腮腺炎暴发的病原学特征及基因变异情况。方法通过Vero/Slam细胞进行病毒分离;采用逆转录-聚合酶链反应(RT-PCR)扩增小疏水蛋白基因片段进行病毒鉴定;应用序列分析软件对病毒基因特征进行比较分析。结果成功分离到9株腮腺炎病毒,均属于G基因型。9株分离株核苷酸序列同源性为99.0%-100%,氨基酸序列同源性为98.0%-100%;与国内其他省流行的F基因型代表株核苷酸与氨基酸差异均〉13.0%;与疫苗株的核苷酸及氨基酸平均差异分别为15.3%和34.5%;同2003年流行于英国的G2亚型病毒代表株核苷酸序列同源性最高(96.8-97.2%)。腮腺炎病毒分离株SH基因的核苷酸存在8个特异性突变位点,其中CNT68、ANT199、TNT271是新发现的突变位点;部分氨基酸保守位点也发生了变化。结论此起暴发是由G基因型腮腺炎病毒引起的。
Objective To analyze pathogenic characteristics and genetic variation of mumps virus isolated from an outbreak in Fujian,2015. Methods Mumps viruses were isolated using vero / slam cells. Small hydrophobic( SH) genes were amplified by reverse transcription-polymerase chain reaction( RT-PCR)and analyzed using software for sequential analysis. Results Nine mumps virus strains were isolated and identified as G genotypes; the homology of nucleotides( nt) and amino acids( aa) among the isolated strains were 99. 0%-100% and 98. 0%-100%,respectively. The difference of nt or aa between isolated strains and predominant F genotype strains circulating in other areas of China was more than 13. 0%. The differences of nt and aa between isolated strains and the S79 vaccine strain were 15. 3% and 34. 5%,respectively. The homology of nt between isolated strains and G2 genotype strains isolated from the United Kingdom in 2003 was 96. 8%-97. 2%. There were eight nt mutations in the SH gene of mumps virus isolates,in which CNT68,ANT199,and TNT271 were newly found,and some conserved aa sites of SH gene also changed. Conclusions This outbreak was caused by the G genotype of mumps virus.
