丙戊酸钠对APP/PS1转基因小鼠脑内海马区老年斑形成的影响

摘要 目的研究丙戊酸钠(VPA)对APP/PS1转基因小鼠脑内海马区老年斑(SP)形成的影响。方法 APP/PS1转基因小鼠随机分为对照组和VPA治疗组,VPA治疗后应用免疫组化技术检测APP/PS1转基因小鼠脑内海马区SP的数量及大小。ELISA测定脑组织β淀粉样蛋白(Aβ)的含量变化。结果 VPA治疗组小鼠海马区域出现的阳性SP数目与对照组相比分别显著减少65%,斑块沉积也分别由0.65%降到0.22%(P<0.01)。ELISA结果显示,VPA治疗组小鼠脑内可溶性Aβ的水平(75 pg/mg)较对照组明显下降(92 pg/ml,P<0.05)。结论 VPA可以减少APP/PS1转基因小鼠脑内SP的形成,抑制Aβ的生成。
