电针对高脂血症新西兰兔脂肪组织脂联素mRNA表达的影响 被引量:2

Effects of electroacupuncture on adiponectin mRNA expression in adipose tissue of New Zealand rabbits with experimental hyperlipidemia

摘要 目的探讨电针胃经穴位对高脂血症新西兰兔血脂及脂肪组织脂联素mRNA表达的影响。方法取24只雄性新西兰兔,随机数字法分为高脂血症组和正常对照组各12只。造模8 w以后,上述两组再各自随机分为电针组和对照组各6只。电针10 d后,7180型日立自动分析仪测定所有新西兰兔的血脂四项,Real Time PCR法检测新西兰兔腹腔脂肪组织脂联素mRNA表达。结果造模后高脂血症组的甘油三酯(TG)、总胆固醇(TC)和低密度脂蛋白胆固醇(LDL-C)明显高于正常对照组,高密度脂蛋白胆固醇(HDL-C)明显低于正常对照组(均P<0.05),显示造模成功。电针后高脂电针组的血清TC水平明显低于高脂对照组(P<0.05)。电针后高脂电针组脂肪组织脂联素mRNA表达量明显高于高脂对照组(P<0.05)。结论电针新西兰兔胃经穴位可以有效降低血脂和增强脂肪组织脂联素基因表达,对改善高脂血症有一定作用。 Objective To investigate the effect of electroacupuncture (EA) on Zusanli and Fenglong acupoint on lipid metabolism and adiponectin mRNA expression in adipose tissue of New Zealand rabbits with experimental hyperlipidemia. Methods 24 clean grade New Zealand rabbits were randomly divided into high lipid group and control group, 12 rabbits in each group. All experimental hyperlipoidemia rabbits were fed by high fat diet for 8 weeks. Then either high lipid group or control group were randomly divided into blank control group and blank EA group, high lipid control group and high lipid EA group,6 rabbits in each group. Rabbits of blank EA group and high lipid EA group were electro-acupunctured for 10 days, while other groups fixed at the same time once per day. Blood was respectively collected from central auricular artery of rabbits before and after modeling, so as before and after treated by EA. All serum was separated. Four items of blood lipid were tested by 7 180 type Hitachi automatic analyzer. Adiponectin mRNA expression in adipose tissue was detected by Real-time PCR. Results (1)The serum levels of TG,TC, LDL-C in rabbits fed by high fat diet were higher than those fed with normal diets ( P〈0.05 ). The serum levels of HDL-C were decreased in rabbits fed by high fat diet were higher than those fed with normal diets( P〈0.05 ). The models were made successfully. (2)With treated by EA for 10 days, the serum levels of TC in rabbits of high lipid EA group decreased significantly (P〈 0. 05 ), compared with high lipid control group. (3)The adiponectin mRNA expression is in adipose tissue of high lipid EA group increased sig- nificantly ( P〈0.05 ) , compared with high lipid control group. Conclusions With EA at Stomach-meridian acupoints, the serum level of TC in hyperlipidemia rabbit is decreased and the adiponectin mRNA expression increased significantly, which can improve symptoms of hyperlipidemia to a certain degree.
