摘要
目的探讨TLR2、mi R-155及SOCS1基因表达在肺结核诊断中的价值。方法采用病例-对照研究设计,纳入研究对象60例,根据肺结核感染状态分为3组:结核阴性对照(Control,n=20)、活动型结核(ATB,n=20)及潜伏型结核(LTB,n=20)。从人全血中分离外周血单个核细胞,提取细胞总RNA,采用实时荧光定量PCR检测TLR2、BIC、mi R-155、SOCS1及相关细胞因子等基因表达情况。结果 Q-PCR结果表明TLR2、SOCS1及mi R-155在各组别中差异表达,进一步统计分析发现,活动型结核组中mi R-155/SOCS1比值显著低于正常组(P<0.05)及潜伏型结核组(P<0.001),而潜伏型结核组中mi R-155/SOCS1比值显著高于正常组(P<0.001)。结论人外周血单个核细胞TLR2、mi R-155及SOCS1的差异表达及mi R-155/SOCS1比值对潜伏性肺结核及活动性肺结核的鉴别诊断具有重要的参考价值。
To investigate the diagnostic value of TLR2,miR-155 and SOCS1 gene expression in tuberculosis(TB),total of 60 subjects were divided into three groups according to their TB state:TB-negative control group(control,n=20),active TB group(ATB,n=20) and latent TB group(LTB,n=20).Firstly,PBMCs were isolated from human blood cells,and then total RNA was extracted from human PBMCs cells using TRIzol reagent.The gene expression levels of TLR2,BIC,miR-155,S0CS1 and related cytokines were detected by real-time PCR.Data showed that there were differential expressions of TLR2,BIC,miR-155 and S0CS1 in PBMCs of patients with active tuberculosis and patients with latent tuberculosis.Further statistical analysis showed that the ratio of miR-155/SOCS1 in ATB group was lower than that in LTB(P〈0.001) and control group(P〈0.05).However,the ratio of miR-155/SOCS1 in LTB group was higher than that in control group(P〈0.001).Our study shows that TLR2,BIC,miR-155,S0CS1 and the ratio of miR-155/SOCSl can be potential biomarkers for discriminating LTB infection and TB disease,which may provide new insights into the diagnosis of tuberculosis.
