摘要
目的:探讨中药复方三棱丸方对大鼠肠纤维化的治疗作用。方法:将48只SD大鼠随机均分为空白组、模型组、罗格列酮对照组、三棱丸方高、中、低剂量治疗组,除空白组外,后五组采每周一次给予5%的TNBS,10 mg于第1天,15 mg于第8天,20 mg于第15天,25 mg于第22天,30 mg于第29天,分别配成50%乙醇溶液0.8 m L一次性灌肠,逐渐增量灌肠法诱导肠纤维化模型;从造模开始,大鼠同时每日分别以10.0 g/kg,5.0 g/kg,2.5 g/kg不同浓度的三棱丸方水煎剂灌胃,罗格列酮组以8 mg/kg浓度罗格列酮灌胃,每天一次;模型组、正常组给予等量生理盐水灌胃。期间观察各组大鼠的一般情况,35 d后收集结肠标本,作病理学评价,检测结肠组织中E-cadherin、α-SMA、FN及PPARγ水平。结果:与模型组比较,三棱丸方高剂量组CMDI评分(P<0.05)与胶原纤维含量均明显低于模型组;E-cadherin、PPARγ蛋白水平均明显增加(P<0.05),FN、α-SMA水平均明显增降低(P<0.05)。结论:三棱丸方能促进PPARγ生成,降低肠道纤维化因子的生成水平,从而产生抗肠纤维化作用。
Objective: To investigate the therapeutic effect of Chinese medicine compound Sparganii Pill on intestinal fibrosis of rats. Methods: Forty-eight SD rats were randomly divided into blank group,model group,rosiglitazone control group,Spargani I Pill high,middle and low dose treatment groups. Except the blank group,the other five groups were given 5% TNBS 10 mg on the first day,15 mg on the 8th day,20 mg on the 15 th day,25 mg on the 22 th day and 30 mg on the 29 th days by enema in 50% ethanol solution 0. 8 m L respectively,which made a TNBS-induced intestinal fibrosis model. Starting from modeling,rats at the same time were given respectively Spargani I Pill with different concentrations( 10. 0 g/kg,5. 0 g/kg,2. 5 g/kg) orally. Rosiglitazone group with rosiglitazone with 8 mg/kg) orally,once a day. Model group and normal group were given the same amount of saline by gavage. After 35 days,the general situation of rats in each group was observed and the colon specimens were collected for pathology evaluation and we detected the levels of E-cadherin,α-SMA,FN and PPARγ. Results: Compared with the model group,CMDI score( P〈0. 05) and collagen fiber content of Sparganii Pill high dose group were significantly lower than those in the model group. E-cadherin and PPARγ protein levels in Sparganii Pill high dose group were significantly increased( P〈0. 05) while FN and α-SMA levels were significantly decreased( P〈0. 05). Conclusion: Sparganii Pill can promote the generation of PPARγ,decrease the level of intestinal fibrotic factor and shows an effect of anti intestinal fibrosis.
