大黄附子成分配伍治疗大鼠阳虚便秘的整合药效学研究 被引量:3

摘要 目的:探讨大黄附子成分配伍后对阳虚便秘大鼠的治疗作用及整合药效学特征。方法:采用山西白醋和0℃冰水活性炭建立大鼠阳虚便秘动物模型,随机分成模型空白组、附子模型组、配伍模型组;将正常大鼠分成正常空白组、附子正常组、配伍正常组,对模型空白组和正常空白组灌胃相应生理盐水,其他4组灌胃相应药物,采血。按照试剂盒说明书测定大鼠不同时间点血浆中胃动素(motillin,MTL)、胃泌素(gastrin,GT)、内皮素(endothelin,ET)、血管活性肠肽(vasoactive intestinal peptide,VIP)的浓度,并进行主成分分析。结果:配伍正常组与附子正常组比较,配伍模型组与附子模型组比较,大鼠各时间点血浆中MTL、GT、ET、VIP各自的浓度和四者的整合浓度均明显升高,且均在180 min时升高最明显,差异有统计学意义(P<0.05或P<0.01)。结论:附子总生物碱和大黄蒽醌配伍后可有效治疗大鼠阳虚便秘;MTL、GT、ET、VIP的整合药效学数据仍然具有明显的药效学特征,且整合后与整合前各成分的药效学特征基本一致,可表达上述4种成分整体的药效学特征。 Objective: To study integrated pharmacodynamics and therapeutic action on constipation rats after the compatibility of total alkaloids from Radix Aconitine and rhubarb anthraquinones. Methods: Shanxi vinegar and 0 ℃ ice water with activated carbon were used to build a constipate rat model. And then,constipation rats were randomly divided into blank model group,Fuzi model group and compatibility model group. But the normal rats were divided into normal blank group,Fuzi normal group and compatibility normal group. The two blank groups were given normal saline via intragastric administration and the other four groups were given corresponding drugs by intragastric administration,respectively. At last,the blood was collected into anticoagulant tubes. According to kit instructions,the concentrations of motillin,gastrin,endothelin and vasoactive intestinal peptide in rats plasma at different time points were measured and the principal component analysis was conducted. Results: The concentrations of MTL,GT,ET,VIP and the integrated concentrations of four ingredients above in rats plasma at different time points in compatibility normal group were higher than those in Fuzi normal group and compatibility model group were also higher than those in Fuzi model group. Both of them reached the highest at 180 min. The difference was statistically significant( P〈0. 05 or P〈0. 01).Conclusion: After the compatibility of total alkaloids from Radix Aconitine and rhubarb anthraquinones,constipation rats should be effectively treated. Significant pharmacodynamic characteristics were still showed by integrated pharmacodynamic data of MTL,GT,ET and VIP. Pharmacodynamic characteristics of components above were consistent before and after the integration. Whole pharmacodynamic characteristics of four components above cloud be expressed by integrated pharmacodynamic characteristics.
