DCE01菌株降解大麻韧皮的发酵液成分变化规律 被引量:2

The Change Law of Hemp Bast Fermentation Broth by Degradation using DCE01 Bacterial Strain

摘要 为阐明草本纤维植物的非纤维素物质生物降解机理,采用高效脱胶菌株DCE01对大麻韧皮进行浸泡振荡发酵,利用平板活菌计数法、DNS法、COD测定法、柱前衍生-高效液相色谱法检测发酵液中生物降解非纤维素物质相关数据的变化趋势。结果表明:发酵液中DCE-01活菌量随发酵时间延长而增加,6.5 h以后的增幅变缓;果胶酶、甘露聚糖酶和木聚糖酶的催化活性在发酵过程中逐渐增大,在10.5 h时最大,依次为411.4 IU/m L、521.4 IU/m L和45.6 IU/m L;COD值以及酶降解产物鼠李糖、葡萄糖醛酸、半乳糖醛酸、半乳糖、木糖等单糖类组分的含量均随发酵时间延长而增加,而甘露糖被DCE01菌株用作碳源,其含量在19.8-23.6μg/m L之间小幅度变化,葡萄糖含量则随发酵时间延长呈现两次先升后降的变化规律。 In order to clarify the mechanism of bio -degumming for bast fiber crop by strain DCE01, the degradation data and change law of the fermentation broth were deeply analyzed by bacteria counting method, DNS method, COD detector, HPLC testing. The results indicated that the living cell concentra- tion increased with the extension of fermentation time, and it had descending growth after 6. 5 h; pecti- nase, mannanase and xylanase activity gradually increased during fermentation, followed by 411.4 IU/ mL, 521.4 IU/mL and 45.6 IU/mL resepectively ; the COD values and the content of enzymatic hydrolysates (rhamnose, glucuronic acid, galacturonic acid, galactose, and xylose) increased with time increasing. Mannose may be used as carbon source by DCE01 strains, and the concentration was 19. 8 - 23.6 μg/mL. The change of glucose levels was presented twice with the rule of rising first and falling later.
