摘要
目的探讨PRKAA1和UNC5CL基因位点多态性与胃癌发生的关系。方法采用基质辅助激光解吸附电离飞行时间质谱检测技术对60例胃炎和60例胃癌患者的rs10074991、rs13361707、rs10036575、rs2294693、rs742494位点进行基因分型。结果PRKAA1基因rs13361707位点CC基因型和○10074991位点GG基因型在胃癌组频率明显高于胃炎组,差异有显著性(P<0.05)。多因素Logistic回归分析校正性别年龄后,差异仍有显著性。rs10036575位点胃癌组CT+CC基因型频率低于胃炎组,差异有显著性(P<0.05)。多因素Logistic回归分析校正性别年龄后,差异仍有显著性。UNC5CL基因rs2294693位点CC基因型和rs742494位点TT基因型频率胃癌组高于胃炎组,但差异无显著性(P>0.05),多因素Logistic回归分析校正性别年龄后,差异无显著性。结论 PRKAA1基因rs13361707位点CC基因型、rs10074991位点GG基因型能增加胃癌的发病风险,而rs10036575位点CT+CC基因型降低胃癌的发病风险。UNC5CL基因rs2294693和rs742494位点基因多态性与胃癌发病风险无关。
Objective To explore the relationship between the polymorphism of PRKAA1 and UNC5 CL and the susceptibility to gastric cancer.Method A total of 120 patients(including 60 gastritis,60 gastric cancer) from Dec.2014 to Dec.2015 matrix-assisted laser desorption/ionization time-of-flight mass spectrometry was used to determine the genotype of rsl0074991,rsl3361707,rsl0036575,rs2294693 and rs742494.x^2 test and logistic regression model were used to analyze the association.Result The frequency of rsl3361707 CC genotype and rsl0074991 GG genotype of PRKAA1 gene in gastric cancer was significantly higher than that in gastritis,the difference was statistically significant(P 0.05).Multivariate logistic regression analysis adjusted for gender and age,the difference was still statistically significant.The frequency of rsl0036575 CC + CT genotype of PRKAA1 gene in gastric cancer was lower than that in gastritis,the difference was statistically significant(P〈0.05).Multivariate logistic regression analysis adjusted for gender and age,the difference was still statistically significant.The frequency of rs2294693 CC genotype and rs742494 TT genotype of UNC5 CL gene in gastric cancer was significantly higher than that in gastritis,but the difference was no statistically significant(P〉0.05).Multivariate logistic regression analysis adjusted for gender and age,the difference was still no statistically significant.Conclusion PRKAAI rs 13361707 CC genotype and rs 10074991 GG genotype increase the risk of gastric cancer,however,rs 10036575 CT+CC genotype decrease the risk of gastric cancer.The polymorphisms of rs2294693 and rs742494 of VNC5CL gene were not associated with the risk of gastric cancer.
