摘要
目的研究雷公藤对正常和肾小管S1段损伤急性肾功能衰竭模型大鼠肾毒性的差异。方法观察雷公藤提取物(TWHF)对生理和病理状态大鼠的一般生长状态、肾脏脏器指数、血清生化指标的影响,并通过病理切片观察大鼠肾脏组织损伤情况。结果 TWHF呈剂量依赖增高的正常大鼠血清尿素氮(BUN)、血清肌酐(CREA)、血清高K^+和低Na^+水平,与其一般生长状态、脏器指数和肾功能损伤病理检查结果相符;低剂量TWHF在模型大鼠上表现出改善BUN、CREA、血清高K^+和低Na^+水平的肾功能保护作用,病理损伤程度也有一定减轻,但高剂量时反而加重模型组大鼠肾功能损伤。结论雷公藤对正常大鼠有剂量相关的肾毒性,但在一定剂量范围内对肾功能损伤病理状态有保护作用,为雷公藤"有故无殒"的安全用药提供了依据。
Objective To study the differences in the renal toxicity of Tripterygium wilfordii Hook. f.( TWHF) on normal and acute renal failure model rats with tubular S1 damaged. Methods The general growth status,organ / body coefficients and serum biochemical indexes were used to investigate the influence of the TWHF on physiological and pathological state. Pathological sections were prepared to observe the damage in renal tissue. Results TWHF increased BUN,CREA,and serum high K + and low Na + levels of normal rats in a dose dependent,which was consistent with its general growth state,organ / body coefficients,and kidney damage pathological examination results. Low doses of TWHF improved the state of BUN,CREA,serum high K + and low Na + levels,and reduced the renal pathological damage in a certain degree,which showed protection effect in model rats. However,high doses of TWHF aggravated kidney injury of model rats. Conclusion Although TWHF has dose related renal toxicity to normal rats,it also has a protective effect on kidney injury pathological status in certain dose range,which provided a basis for clinical safe application of TWHF based on symptom- based prescription theory.
