摘要
目的探讨欧芹根总黄酮对非酒精性脂肪性肝病(NAFLD)大鼠模型的影响机制。方法采用高脂饮食诱导NAFLD大鼠40例建模,随机分为正常组、模型组、水飞蓟素组和欧芹根总黄酮组,各10只,分别给予生理盐水、生理盐水、水飞蓟素、欧芹根提取物,除正常组外,其他组均每日喂食高脂饲料。检测各组大鼠血清中丙氨酸氨基转移酶(ALT)、天门冬酸氨基转移酶(AST)、总胆固醇(TC)、三酰甘油(TG)水平,采用实时荧光定量聚合酶链反应(Quantitative Real-time PCR)法检测肝组织中环氧合酶-2(COX-2)的mRNA表达水平。结果建模成功,模型组大鼠血清ALT,AST,TC,TG水平与正常组大鼠相比均明显升高(P<0.05);水飞蓟素和欧芹根总黄酮干预后能显著降低NAFLD大鼠上述4项指标水平,并抑制COX-2的mRNA表达,差异均有统计学意义(P<0.05);欧芹根总黄酮作用略强于水飞蓟素,但差异无统计学意义(P>0.05)。结论欧芹根总黄酮对NAFLD大鼠有一定的治疗作用,其作用机制与抑制肝组织中COX-2的表达有关。
Objective To explore the effect mechanism of total flavonoids in roots of petroselinum crispum on nonalcoholic fatty liver rat model. Methods 40 cases of nonalcoholic fatty liver rat model were induced by high- fat diet and then randomly divided into the normal group,model group,silymarin group and total flavonoids in roots of petroselinum crispum group,10 rats in each group. All group were given the corresponding treatment. Except for the normal group,the other groups were fed with high- fat diet. The plasma level of ALT,AST,TG,TC were detected and the expression of COX- 2 mRNA in liver was examined by Quantitative Real- time PCR. Results The model was made successfully,and the plasma level of ALT,AST,TG,TC in model groups were higher than that in normal group. After intervention,the NAFLD rats with silymarin and total flavonoids in roots of petroselinum crispum decresed the above- mentioned four indexes obviously,and the expression of COX- 2 mRNA was inhibited( P〈0. 05). The effects of total flavonoids in roots of petroselinum crispum were stronger than silymarin,but the differences were not statistically significant( P〈0. 05). Conclusion The total flavonoids in roots of petroselinum crispum has a specific curative effect on NAFLD rats. Its mechanism is related to the inhibition of COX- 2 mRNA expression.
