摘要
目的检测survivin和SIRT1 mRNA在非小细胞肺癌中的表达,探讨其相互关系。方法用实时定量PCR的方法检测江苏省肿瘤医院42组非小细胞肺癌组织和对应的癌旁组织中survivin和SIRT1 mRNA的表达,统计survivin与SIRT1的相关性。观察EGFR酪氨酸酶抑制剂吉非替尼处理非小细胞肺癌细胞系HCC827后survivin和SIRT1的表达改变。结果 85.7%(36/42)肿瘤组织survivin mRNA表达明显高于对应的癌旁组织。61.9%(26/42)肿瘤组织SIRT1 mRNA的表达低于其癌旁组织,33.3%(14/42)肿瘤组织SIRT1 mRNA表达高于其癌旁组织。Survivin表达与SIRT1表达有统计学意义的相关性(P<0.05)。吉非替尼能增加HCC827细胞中的SIRT1表达,降低survivin的表达水平,呈时间和剂量依赖性。结论 Survivin在非小细胞肺癌组织中高表达,而大部分SIRT1基因呈低表达状态。survivin与SIRT1表达呈负相关。在细胞水平上,吉非替尼能增加SIRT1的表达从而降低survivin的水平。提示两者在非小细胞肺癌的发生发展中可能发挥重要作用。
Objective The aim of the study is to detect the expression of survivin and SIRT1 in non-small cell lung cancer and their correlation. Methods The expression of survivin mRNA and SIRT1 mRNA was detected by real-time fluorescence quantitative PCR. Results 85.7% of Survivin mRNA expression in tumor tissues was higher than that in their adjacent mucosa. 61.9% of SIRT1 mRNA expression in tumor tissues was lower than that in their adjacent mucosa while 33.3% was elevated in tumor tissues. Survivin expression was closely correlated with SIRT1 expression. Epidermal growth factor receptor tyrosine kinase inhibitor, gefitinib, increased SIRT1 expression and decreased survivin expression in a dose and time-dependent manner. Conclusions The expression of survivin in non-small cell lung cancer is higher than that in adjacent mucosa tissues while SIRT1 expression is lower. There is a relationship between expression of survivin and SIRT1.
