The effects of Caveolin-1 on migration and invasion of breast cancer

摘要 目的探讨Caveolin-1在乳腺癌组织中的表达,分析过表达Caveolin-1对乳腺癌细胞增殖、迁移及侵袭的影响。方法qRT-PCR、Westernblot法检测乳腺癌组织及细胞中Caveohn-1mRNA及蛋白的表达情况:MTY、Transwell实验检测质粒过表达Caveolin-1表达后对乳腺癌细胞增殖、侵袭及迁移的影响。结果乳腺癌组织Caveolin-1 mRNA转录水平及蛋白表达水平均显著低于癌旁组织(P〈0.05);Caveolin-1mRNA及蛋白在乳腺癌细胞MDA-MB-231及MCF7细胞中明显低于SKBR3、BT474(P〈0.05)。转染质粒可成功上调Caveolin-1在MDA-MB-231及MCF7中的表达.经质粒Caveolin-1过表达后乳腺癌细胞的增殖能力明显下降(P〈0.05)、迁移能力和侵袭能力也明显下降(P〈0.05)。结论Caveolin-1在乳腺癌组织中的表达明显降低,并抑制乳腺癌细胞增殖、侵袭及迁移。 Objective To investigate the expression of Caveolin-1 in breast cancer patients and to ex- plore its role in proliferation, migration and invasion of breast cancer cells. Methods The expression of Cave- olin-1 in breast cancer tissues and corresponding adjacent noncancerous tissue was assessed by RT-PCR and Western blot. The role of Caveolin-1 in proliferation, invasion and migration of breast cancer cell MCF7 and MDA-MB-231 was observed by plasmid overexpression, MTr and Transwell assay. Results The Caveolin-1 mR- NA transcription and protein expression in breast cancer tissue were significantly lower than tissue adjacent to carcinoma (P〈0.05); Caveolin-1 mRNA and protein in breast cancer cells MDA-MB-231 and MCF7 were signifi cantly lower than SKBR3, BT474 cells (P〈O.05). The overexpression of Caveolin-1 by plasmid could inhibit the prolif- eration, invasion and migration of MCF7 and MDA-MB-231 by reducing MMP2 and MMP9. Conclusion Caveolin-1 expression is down regulated in breast cancer tissue and this contributes to the proliferation, migration and invasion of breast cancer by upregulating MMP2 and MMP9.
