高效液相色谱法测定三七药材中硝酸盐含量

Determination of Nitrate Content in Panax Notoginseng by HPLC

采用高效液相色谱法测定了三七中硝酸盐（NO-3）的含量。Utimate pheny1-ether（150 mm×4.6 mm,5μm）为色谱柱,以四丁基溴化铵（TBABr）作为离子对试剂,流动相为乙腈-0.05mol/L磷酸二氢钾（KH2PO4）溶液＋0.03 mmol/L四丁基溴化铵（用磷酸调节pH值至3.3±0.1）=10∶90;柱温30℃,流速1.0 m L/min,检测波长216 nm,进样量10μL。结果显示：三七药材中硝酸盐在5~500μg范围内呈良好的线性关系（r=0.999）,平均回收率为100.2%（n=9）,RSD=1.6%（n=9）。本方法简便、快速,结果准确、可靠、重现性好。适用于三七药材中硝酸盐的含量测定。

The determination of the nitrate content in Panax notoginseng by HPLC was discussed. The separation was performed with tetrabutyl ammonium（ TBABr） dissolved in acetonitrile-0. 05 mol/L monopotassium phosphate（ KH2PO4）（ 10∶90） buffer as mobile phase at a flow rate of 1. 0 m L/min and accomplished with utimate phenyl-ether（ 150 mm×4. 6 mm,5 μm） column at 30 ℃. The detection wavelength was 216 nm and the injection volume was 10 μL.Under the detection conditions,the nitrate content in Panax notoginseng showed good linearity（ r = 0. 9998）. The recovery rate was 100. 2%（ n = 9）,RSD = 1. 6%（ n = 9）. This method is simple,rapid,accurate,reliable and reproducible. It is suitable for detecting various contents of nitrate in Panax notoginseng.
