高效液相色谱法测定三七药材中硝酸盐含量

摘要 采用高效液相色谱法测定了三七中硝酸盐(NO-3)的含量。Utimate pheny1-ether(150 mm×4.6 mm,5μm)为色谱柱,以四丁基溴化铵(TBABr)作为离子对试剂,流动相为乙腈-0.05mol/L磷酸二氢钾(KH2PO4)溶液+0.03 mmol/L四丁基溴化铵(用磷酸调节pH值至3.3±0.1)=10∶90;柱温30℃,流速1.0 m L/min,检测波长216 nm,进样量10μL。结果显示:三七药材中硝酸盐在5~500μg范围内呈良好的线性关系(r=0.999),平均回收率为100.2%(n=9),RSD=1.6%(n=9)。本方法简便、快速,结果准确、可靠、重现性好。适用于三七药材中硝酸盐的含量测定。 The determination of the nitrate content in Panax notoginseng by HPLC was discussed. The separation was performed with tetrabutyl ammonium( TBABr) dissolved in acetonitrile-0. 05 mol/L monopotassium phosphate( KH2PO4)( 10∶90) buffer as mobile phase at a flow rate of 1. 0 m L/min and accomplished with utimate phenyl-ether( 150 mm×4. 6 mm,5 μm) column at 30 ℃. The detection wavelength was 216 nm and the injection volume was 10 μL.Under the detection conditions,the nitrate content in Panax notoginseng showed good linearity( r = 0. 9998). The recovery rate was 100. 2%( n = 9),RSD = 1. 6%( n = 9). This method is simple,rapid,accurate,reliable and reproducible. It is suitable for detecting various contents of nitrate in Panax notoginseng.
