沉默干扰素诱导蛋白16对人主动脉外膜成纤维细胞凋亡的影响

摘要 目的探究沉默干扰素诱导蛋白16(IFI16)对人主动脉外膜成纤维细胞(HAAFs)凋亡的影响。方法检测HAAFs凋亡、IFI16、细胞外调节蛋白激酶1/2(ERK1/2)及磷酸化细胞外调节蛋白激酶1/2(p-ERK1/2)蛋白表达。结果基因沉默组IFI16低于另两组(P<0.05),基因沉默组ERK1/2及p-ERK1/2蛋白表达高于另两组(P<0.05),阴性对照组及空白组HAAFs凋亡率、IFI16、ERK1/2及p-ERK1/2蛋白表达差异无统计学意义(P>0.05)。结论沉默IFI16表达能够降低HAAFs凋亡率。 Objective To explore the effect of silencing interferon inducible protein 16(IFI16)on the apoptosis of human aortic outer membrane fibroblasts(HAAFs). Methods Detection of HAAFs apoptosis,IFI16 and extracellular regulated protein kinase 1/2(ERK1/2)and phosphorylated extracellular regulated protein kinase 1/2(p-ERK1/2)protein expression. Results Gene silencing of group IFI16 was lower than that of the other two groups(P 〈0.05),gene silencing and p-ERK1/2 protein expression in ERK1/2 group was significantly higher than that of the other two groups(P 〈0.05),negative control group and blank control group HAAFs,IFI16,ERK1/2 apoptosis and p-ERK1/2 protein expression differences were not statistically significant(P 〉0.05). Conclusion Silencing IFI16 expression can reduce the apoptosis rate of HAAFs.
