中国首例黄热病输入病例系列样本的实验室检测报告 被引量:4

Report of the laboratory detection of the series samples from the first imported yellow fever case in China

摘要 目的观察中国首例输入性黄热病病例不同时间和不同类型样本中病毒核酸检测结果对疾病诊断的意义。方法采用实时荧光RT-PCR法,对不同时间采集的不同类型样本进行黄热病毒核酸检测。结果不同时间采集的血清样本中,病毒核酸的量随病程延长而降低,Ct值依次为发病3d时25.11,4d时26.00,7d时28.84。不同类型样本中可检测到的病毒核酸量不同,发病3d时,血清病毒核酸检测的Ct值为25.11,血块为25.80;发病4d时,血清病毒核酸检测的Ct值为26.00,血块为28.80,唾液为35.00;发病7d时,血清病毒核酸检测的Ct值为28.84,血块为27.38。结论黄热病感染早期,病毒核酸为灵敏、特异的实验室检测指标;综合考虑样本采集以及前处理的难易程度等因素,血清样本可作为黄热病早期病毒核酸检测适宜的临床样本,其检测结果对疾病诊断具有重要意义。 Objective To investigate the significance of viral RNA detection in different type of samples from different time for the diagnosis of yellow fever based on the first imported case. Methods Yellow fever virus RNA was detected by real-time RT-PCR in different type of samples from different time. Results In the ser- um samples collected from different time, the quantity of viral RNA decreased with the time course of the dis- ease, the Ct value was 25.11 on the 3 d, 26.00 on the 4 d and 28.84 on the 7 d, respectively. In different type of samples, the quantity of viral RNA was also different. The Ct value was 25.11 in serum and 25.80 in blood clot on the 3 d;26.00 in serum, 28.80 in blood clot and 35.00 in saliva on the 4 d; 28.84 in serum and 27.38 in blood clot on the 7 d. Conclusions In the early phase of yellow fever, viral RNA is a sensitive and specific marker for virus detection. Taking all factors into consideration, such as easily collecting and sample prepro- cessing, serum is the appropriate clinic sample for the viral RNA detection, and the detection result is signifi- cant for the diagnosis of this disease.
