EDC/NHS法制备免疫磁珠及其效果验证 被引量:3

摘要 免疫磁珠又叫免疫磁性微球,是免疫学和磁性微球结合而发展的一类新型材料,一般用于物质的提取和分离,由于其高效、低毒等特性,目前在生物化学领域、医药领域、食品检测领域应用广泛。本文主要应用EDC/NHS法活化羧基磁珠,再将人类精子特异性抗体与之偶联,最后制备成免疫磁珠,进行细胞捕获实验,分别捕获人类的上皮细胞和精子,提取DNA,进行PCR(聚合酶链式反应),进行电泳,根据STR(短串联重复)图谱的对比验证了这种方法制备的免疫磁珠的偶联效果。实验证实,用EDC/NHS法活化制备的免疫磁珠能选择性地捕获到精子而不能捕获到上皮细胞,该方法制备磁珠效果良好。
