SGK3与浸润性乳腺癌临床病理相关性实验研究

Correlation study between protein kinase SGK3 and clinical pathological in breast cancer

摘要 目的通过浸润性乳腺癌中血清和糖皮质激素调节蛋白激酶3(serum and glucocorticoid-regulated protein kinase 3,SGK3)的表达情况进行分析,初步探讨SGK3与浸润性乳腺癌临床病理特征的相关性。方法课题组于2015年10月—2016年1月,利用组织芯片,检测SGK3在癌旁乳腺组织和浸润性乳腺癌组织中的表达,并结合临床参数探讨SGK3与浸润性乳腺癌临床病理特征的关系,计量资料比较采用t检验,P<0.05为差异有统计学意义。结果免疫组织化学(组化)结果显示,SGK3在浸润性乳腺癌中的表达高于癌旁正常乳腺组织[(338.157±133.349)、(199.959±52.917)],对比差异有统计学意义(P<0.05),且在浸润性乳腺癌组织中,雌激素受体(estrogen receptor,ER)阳性组织中SGK3的表达较ER阴性组织增高[(378.494±142.749)、(318.853±143.199)],对比差异有统计学意义(P<0.05)。结论 SGK3在浸润性乳腺癌组织中呈高表达,且在ER阳性的浸润性乳腺癌组织中的表达高于ER阴性组织,SGK3有可能成为浸润性乳腺癌临床诊断和预后的指标。 Objective Through the analysis of the expression of serum and glucocorticoid-regulated kinase 3(SGK3) in invasive breast cancer,the correlation between SGK3 and clinical pathological characteristics of invasive breast cancer was preliminarily discussed.Methods During the period from October 2015 to January 2016,tissue microarray technique was used to detect the expression of SGK3 in normal breast tissues and invasive ductal carcinoma tissues.Discussed the relationship between SGK3 and clinical pathological characteristics of invasive breast cancer was in combination with clinical parameters.The measurment data comparison between the two groups use t-test,P〈0.05 indicated that the difference was statistically significant.Results Immunohistochemical results show,the expression of SGK3 in invasive ductal carcinoma was higher than that in adjacent normal breast tissues[(338.157±133.349)、(199.959±52.917)](P〈0.05).The expression of SGK3 in the invasive ductal carcinoma tissues of the estrogen receptor positive was significantly increased.Compared with that of the estrogen receptor negative[(378.494±142.749)、(318.853±143.199)],there were significant differences(P〈0.05).Conclusion SGK3 was highly expressed in breast invasive ductal carcinoma tissues.It was positively correlated with the pathological grade of tumor and estrogen receptor expression.SGK3 may be a target for clinical diagnosis and prognosis of breast invasive ductal carcinoma.
