摘要
目的探讨蛋白酪氨酸磷酸酶IB(PTP1B)在乳腺非特殊型浸润性癌(NST)组织中的表达及意义。方法采用原位杂交技术和免疫组化法分别检测86例乳腺NST组织中PTP1BmRNA和蛋白的表达,并分析其与乳腺癌临床病理特征的关系,以癌旁正常乳腺组织作为阴性对照。结果PTP1BmRNA及蛋白在乳腺癌组织中的阳性表达率分别为52.3%和59.3%,均明显高于癌旁正常乳腺组织,差异有统计学意义(P≤0.01)。PTP1BmRNA与蛋白表达呈正相关(rs=0.773,P〈0.001),且PTP1B表达与乳腺癌淋巴结转移、TNM分期有关(P≤0.001),与者患者年龄、肿瘤大小和组织学分级无关(P均〉0.05)。生存分析结果显示,PTP1B蛋白阳性患者的生存时间短于阴性患者(x2=13.071,P〈0.001)。结论PTP1B在乳腺癌组织中高表达,且与淋巴结转移、TNM分期有关,其有望成为乳腺癌治疗的潜在靶点。
Objective To explore the expression and significance of protein tyrosine phosphatase 1B ( PTP1 B) in invasive breast canc- er of no special type (NST) . Methods In situ hybridization technique and immunohistochemical staining were used to detect the expres- sions of PTP1B mRNA and protein in 86 NST breast cancer patients, the relationships with clinicopathological parameters were analyzed, the normal breast tissues adjacent to breast cancer were selected as negative control group. Results The positive expression rates of PTP1B mR- NA and protein in breast cancer were 52. 3% and 59. 3%, respectively, which were statistically significantly higher than those in normal breast tissue (P≤0.01) . There was a positive correlation between the expression of PTP1B mRNA and protein (rs =0. 773, P〈0. 001 ) . PTPiB: expression was significantly correlated with lymph node metastasis and TNM stage (P≤0. 0Q1 ), which was not correlated with age, tumor size and histological grade (P〉0. 05) . Surival analysis showed that the survival time of PTP1B protein positive patients was shorter than that of PTP1B protein negative patients (x2 = 13.071, P〈0. 001 ) . Conclusion The expression of PTP1B is up-regulated in breast cancer, which is correlated with lymph node metastasis and TNM stage, PTP1B may be a potential therapy target for breast cancer.
