摘要
目的:观察龟羚帕安丸对帕金森病(PD)大鼠病理学、行为学及中脑黑质突触核蛋白(SYN)、Caspase-3表达的影响。方法:采用6-OHDA注射法建立PD大鼠模型,造模成功大鼠随机分为模型组、美多芭组、龟羚帕安丸大、中、小剂量组(4、2、1 g·kg^(-1)),同时设立假手术组,假手术组、模型组给予等容积生理盐水,连续灌胃给药4周。观察各组大鼠旋转行为、中脑黑质细胞病理学及Caspase-3、SYN表达情况。结果:与假手术组比较,模型组大鼠神经细胞变性坏死明显增多,旋转圈数多,SYN、Caspase-3表达明显升高(P<0.01);各用药组大鼠旋转圈数显著低于模型组,SYN、Caspase-3表达显著低于模型组(均P<0.01)。结论:龟羚帕安丸可以改善帕金森病模型大鼠病理改变及行为学,下调Caspase-3、SYN表达可能是其治疗帕金森病的机制之一。
Objective : To observe the efficacy of Guiling Pa'an Pill on Parkinson' s disease (PD) rats' pathology, be- havior and substantia nigra of synuclein (SYN) and Caspase - 3. Method : The rats were injected with 6 - OHDA solution into the left substantia nigra compact part. Rats showed consistently right whirling and the number of rotation was more than 210 r· 30 min-1 induced by APO,and then the rats were judged as the PD model. Fifty model rats were randomly divided into five groups:model group,madopar group( 10 mg · kg-1 ) ,Guiling Pa' an Pill group( 1.0 g· kg-1 ,2.0 g · kg-1 ,4.0 g ·kg-1 ). At the same time,the sham operation group was established and given normal saline with the same volume, all groups with a course of 30 days. The cell pathology, rotation behavior and synuclein protein levels of SYN and Caspase - 3 were detected. Result : The nerve cell degeneration in model group was more than that of the other groups sig- nificantly. The number of rotations was significantly higher than that of the other groups. The synuclein proteins in SYN and Caspase- 3 were significantly increased in model group( P 〈 0.01 ). Compared with model group,the synuclein pro- teins in SYN and Caspase - 3 were significantly decreased in madopar group and Guiling Pa'an Pill groups. The number of rotations was significantly decreased(P 〈 0. 01 ). Conclusion: Guiling Pa'an Pill can improve pathology and behavior of rats model of Parkinson's disease and the down - regulation of SYN and Caspase - 3 expressions may be one of its mecha- nisms for the treatment of Parkinson's disease.
