半夏泻心汤拆方对胃溃疡大鼠胃组织GAS、MTL与血清PAF、ET、NO的影响及其寒热并用配伍的意义研究 被引量:22

摘要 目的:通过观察半夏泻心汤拆方各组对胃溃疡大鼠模型的胃组织中GAS、MTL及血清中PAF、ET含量变化的影响,分析本方治疗胃溃疡的作用机制,探讨该方中不同药物组在治疗胃溃疡中的配伍意义,揭示寒热并用配伍方法的优势及作用靶点。方法:选取SD大鼠,雌雄各半,先用苦寒泻下法制作脾胃虚寒模型,再用辣椒与乙醇溶液,配合乙酸胃黏膜注射建立胃溃疡寒热错杂证-病证结合动物模型。胃溃疡造模24 h后,各组灌胃给药,分别给予半夏泻心汤及拆方各组药物,第28天处死各组大鼠,采集适量胃组织和血清,运用酶联免疫吸附实验检测胃组织中GAS、MTL及血清中PAF、ET含量,硝酸还原酶法测定血清中NO的含量;结果:与KB组比较,MX组大鼠胃组织中GAS、MTL及血清中PAF、ET含量显著升高,血清中NO含量显著降低,差异有统计学意义(P<0.01);与MX组比较,QF组、HR组、HB组及YX组胃组织中GAS、MTL含量显著降低(P<0.01),QF组、HR组、WR组及WB组的PAF、ET含量显著降低(P<0.01),HR组、WR组、BY组、WB组、HB组及YX组的NO含量显著升高(P<0.01);与QF组比较,HR组和YX组的GAS含量无显著性差异(P>0.05),HR组、HL组和HB组的MTL含量无显著性差异(P>0.05);HR组的PAF含量无差异(P>0.05),HR组及WR组的ET含量无差异(P>0.05),HR组、WR组和WB组的NO含量无差异(P>0.05)。结论:半夏泻心汤方中HL组及HB组在降低胃组织中GAS、MTL的含量方面作用显著。WR组在升高血清中NO含量,降低血清中PAF、ET含量方面作用显著。寒热并用配伍能够消除单独使用HL组或WR组药物的局限和不足,通过不同的途径和作用靶点来发挥对胃溃疡的治疗作用,并且在多个方面产生了协同增效作用。 Objective : To discuss the influence of Banxia Xiexin Decoction and different components formula on gastric ulcer rats' GAS and MTL in gastric tissue and PAF, ET and NO contents in serum and analyze the mechanism and its subdivisions to reveal the superiority and targets of both cold and heat compatibility.Methods : Select male and female SD rats and establish the gastric ulcer model of mixed cold and heat syndromes. After 24 hours, all the rats except those of the blank group and sham operation group were given Banxia Xiexin Decoction and different components formula. All rats were sacrificed after 28 days. Observe the rats'pathological morphology change of gastric tissue and test NO content in serum by nitrate reductase method. And the TNF-α,IL -8,GAS and MTL in gastric tissue and PAF and ET contents in serum were detected by ELISA method. Result: Compared with blank group,the contents of GAS, MTL and PAF and ET in serum were significantly elevated in model group( P 〈0.01 ) while the content of NO in serum decreased obviously(P 〈 0.01 ). Compared with model group, the contents of GAS and MTL were significantly decreased in Banxia Xiexin De- coction group,both cold and heat compatibility group, cold tonic group and positive group (P 〈 0.01 ). The contents of PAF and ET in serum were significantly decreased in Banxia Xiexin Decoction group, both cold and heat compatibility group,heat group and heat tonic group(P 〈 0.01 ) while the content of NO in serum decreased obviously( P 〈 0.01 ). Compared with Banxia Xiexin Decoction group, the content of GAS was significantly decreased in both cold and heat corn-patibility group and positive group ( P 〉0. 05 ). The content of MTL was significantly decreased in both cold and heat compatibility group, cold group and cold tonic group( P 〉 0.05 ). The content of PAF in both cold and heat compatibility group had no significant difference ( P 〉 0. 05 ). The content of ET in both cold and heat compatibility group and heat group had no significant difference(P 〉0.05). The content of NO in both cold and heat compatibility group,heat group and heat tonic group had no significant difference( P 〉 0.05 ). Conclusion: The different properties drug Of Banxia Xiexin Decoction plays a therapeutic role from different aspects on gastric ulcer. Cold group drugs and cold tonic group play an important role on reducing GAS and MTL. Heat group drugs play an important role on increasing the content of NO and reducing PAF and ET. After compatibility of both cold and heat drugs, the synergistic effect was reinforced. Compatibility of cold and heat drugs can reduce the limitations and deficiencies of cold or heat drugs and embodies the Chinese medi- cine therapeutic advantage.
