电针对脑出血大鼠海马区Nogo-A表达的影响 被引量:2

摘要 目的探讨电针对脑出血大鼠出血同侧海马区Nogo-A表达的影响及作用机制。方法将66只成年SD大鼠按随机数字表法分为对照组和脑出血组,脑出血组再随机分为非干预组和电针组,Ⅶ型胶原酶脑内注射诱导大鼠尾状核出血,电针组于造模次日开始电针干预,各组大鼠于第2、5、10、15天取出血侧海马C1区脑组织石蜡切片行Nogo-A免疫组化检测,每例脑组织取3张切片,每张切片于400倍光镜下随机选取5个互不叠加视野计数阳性细胞并取其平均值。结果脑出血组大鼠右侧尾状核区域有血肿形成;与对照组比较,非干预组与电针组海马区Nogo-A阳性表达水平均显著增强(P<0.01),而电针组较非干预组表达水平明显下降(P<0.01)。结论电针干预可显著抑制脑出血大鼠脑内Nogo-A的表达,有利于脑出血后中枢神经轴突的再生修复,促进神经功能恢复。 Objective To investigate the effects and mechanism of electro'axupuncture on the expression of Nogo-A in hippocampus at the same side of bleeding in rats with cerebral hemorrhage. Methods Sixty-six adult SD rats were randomly divided into control group and cerebral hemorrhage group by the random number table, and the cerebral hemorrhage group was randomly divided into non-intervention group and electro-axupuncture group. Hemorrhage of caudate nucleus in rats was induced by intracerebral injection of collagenase Ⅶ, the electro-axupuncture group was given eleetro-axupuncture intervention in the next day of molding, the rats of all groups were taken paraffin section of brain tissues in C1 area of hippocampus of bleeding side at the second, fifth, tenth, fifteenth day, each case of brain tissue was taken 3 slices, 5 non-overlapping field of views were randomly selected from each slice under 400 times light micro- scope to count the positive cells and take the average value. Results The cerebral hemorrhage group had hematoma at the right caudate nt^cleus. Compared with the control group, the positive expression levels of Nogo-A in hippocampus of non-intervention group and electro-axupuncture group were significantly enhanced (P 〈 0.01), while the expression level of electro-axupuncture group was significantly lower than that of non-intervention group (P 〈 0.01). Conclusion Electro-axupuncture can significantly inhibit the expression of Nogo-A in rats with cerebral hemorrhage, which is helpful for the regeneration and repair of nervous centralis axon after cerebral hemorrhage and to promote the recovery of neu-rological function.
