旋复代赭汤及其拆方对反流性食管炎模型大鼠食管线粒体UCP2表达的影响 被引量:12

Effects of Xuanfu Daizhe Decoction and Its Decomposed Prescription on UCP2 Expression in Esophageal Mitochondrion of Rat Model with Reflux Esophagitis( RE) Disease

摘要 目的:本实验旨在通过分析旋复代赭汤及其拆方对反流性食管炎(RE)模型大鼠食管组织线粒体UCP2表达的影响,探讨气机升降与线粒体能量代谢调控机制的相关性,以期进一步阐释RE的病机及旋复代赭汤治疗RE的作用机理。方法:将150只健康雄性Wister大鼠随机分为10组,即对照组(正常组、模型组)、旋复代赭汤全方组及拆方组(苦降组、甘升组、升降相因组、全方去苦降组、全方去甘升组、全方去升降相因组)与西药组,每组15只。采用"4.2mm幽门夹+胃底2/3结扎术"制备酸碱混合反流性食管炎大鼠模型。术后第8天开始,旋复代赭汤全方组及拆方各组分别给予相应药液灌胃,西药组给予兰索拉唑+莫沙必利灌胃,对照组(正常组+模型组)均给予等量生理盐水灌胃。干预14天后,第22天处死所有模型大鼠。在光镜下观察食管黏膜组织病理变化,采用Western-blot法检测各组RE模型大鼠食管组织线粒体中UCP2的相对表达量。结果:通过Western-blot相对灰度的比较分析,正常组UCP2的相对表达量最低,同全方组及西药组比较无明显差异(P>0.05);而全方组及西药组UCP2的相对表达量明显低于模型组(P<0.01);全方去苦降组、甘升组、升降相因组与模型组比较,结果有差异(P<0.05);其余各组(全方去甘升组、全方去升降相因组、苦降组)UCP2的相对表达量与模型组比较无明显差异(P>0.05),不具有统计学意义。结论:旋复代赭汤可以明显降低RE大鼠食管组织线粒体UCP2的表达量,表明此方能够很好的调节RE大鼠食管线粒体能量代谢,以此推断该方能明显改善RE大鼠食管下端括约肌松弛,在缓解胃食管反流方面具有良好的疗效。 Objective: The aim of this article was to discuss the correlation of Qi ascending and descending the of theory and the energy metabolism regulatory mechanism,to further explain the mechanism that the prescription affect on RE and the pathogenesis of RE,by analyzing the affection that the prescription of Xuanfu Daizhe Decoction and those divided groups had on the reflux esophagitis( RE) of the rats model esophageal tissue mitochondrion membrane potential UCP2. Methods: 150 healthy male Wister rats were randomly divided into 10 groups,namely,controlled group( model controlled group,normal controlled group),Xuanfu Daizhe Decoction all partys group and divided groups( bitter descending group,and umami ascending group,ascending and descending group,all partys excluding descending group,all partys excluding ascending and descending group),and western medicine group,each group was 15 ones. The rats model of RE with "4. 2mm pylorus clip + ligation of stomach bottom two-thirds",to prepare to mix acid and alkali. The whole group( Xuanfu Daizhe Decoction) and divided group were filled the stomachs with corresponding solution respectively,while the western medicine group was given( mosapride + lansoprazole) and the controlled groups( normal group + model group) were given equal saline the 8th days after operation. All the rats would be killed in 22 th day after continued for 14 days intervention. Observing the changes of the esophageal mucosal morphology with optical microscope,and using western blot method to test UCP2 relative expression of each RE model esophageal tissue mitochondria. Results: By comparative analyzing western blot relative gray,the UCP2 relative expression was the lowest in the normal group,there was no statistically difference compared to the whole group and the western medicine group( P〉0. 05); while the UCP2 relative expression of the whole group was lower obviously than western medicine group( P〈 0. 01); the UCP2 relative expression of all partys excluding descending group,umami ascending group,ascending and descending group were statistically different( P〈0. 05),but the others weren't( P〉0. 05),when compared with the model group. Conclusion: Xuanfu Daizhe Decoction is able to descend obviously the UCP2 expression of the RE rats esophageal tissue mitochondriall,which shows that the prescription does well in regulating RE rats esophageal mitochondrial energy metabolism,deducing from that it can significantly improve the lower esophageal sphincter of the RE rats,and has good curative effect on easing RE.
