摘要
目的探讨黄精地龙方(RPEWM)对体外培养分化的SH-SY5Y细胞Aβ_(25-35)损伤中的细胞钙调节相关酶活性及游离钙离子(Ca^(2+))浓度的影响。方法 SH-SY5Y细胞经维甲酸诱导分化,再经Aβ_(25-35)造成细胞老年痴呆(AD)损伤,在细胞AD损伤同时加入RPEWM提取液保护。观察该方对损伤后的细胞增殖,细胞凋亡,细胞Ca^(2+)-ATPase、Ca^(2+)-Mg^(2+)-ATPase活性及游离钙离子浓度的影响。结果 RPEWM提取液可明显抗细胞AD损伤、抗调亡,促进细胞增殖;能升高细胞内Ca^(2+)-ATPase、Ca^(2+)-Mg^(2+)-ATPase活性,降低Ca^(2+)浓度,且呈剂量依赖关系。结论 RPEWM对分化的SH-SY5Y细胞Aβ_(25-35)损伤有良好的保护作用,对AD有一定治疗效果。
Objective To observe the effects of Huangjing Dilong mixtures( HDM) on calcium related enzymes and Ca^2+regulative functions in the damage of Aβ25-35 to the differentiated SH-SY5 Y cells. Methods The SH-SY5 Y cells were differentiated to neuron by ATRA,then impaired by Aβ25-35 as the Alzheimer's disease( AD) injury to cells,at the same time,the extractive of HDM were added to cells act as protector. Finally,the survival rate,apoptotic rate were observed. The Ca^2+-ATPase,Ca^2+-Mg^2+-ATPase activities were detected. The intracellular calcium levels [Ca^2+]i in the AD model's cells were observed. Results Compared with model group,the extractive of HDM could significantly anti-AD impairment to neuron cells,increased the cell viability,survival rate,and decreased the apoptotic rate. Also,HDM could promote Ca^2+-ATPase,Ca^2+-Mg^2+-ATPase activities,and decrease the [Ca^2+]i in the AD model's cells. Conclusion HDM has the function of anti-AD,can effectively protect the differentiated SH-SY5 Y cells against AD impairment which exerted by Aβ25-35.
