摘要
目的:研究雪菊黄酮对发生脂肪变的正常人肝细胞系L-02的影响。方法:医用脂肪乳注射液制备人肝L-02细胞的高脂细胞模型。以不同浓度(20、50、100μmol/L)的雪菊黄酮进行共培养,分别孵育高脂变性的肝细胞不同时间(3、12、24、48 h)后,用油红脂肪染色法观察细胞脂滴形成情况。检测细胞培养上清内的酶学指标谷丙转氨酶(ALT)、谷草转氨酶(AST)、γ-谷氨酰转肽酶(γ-GT)及甘油三酯(TG)的含量。结果:医用脂肪乳培育人肝L-02细胞脂肪变性模型可行。高浓度的雪菊黄酮(100μmol/L)培养后高脂肝细胞胞浆内脂滴明显减少,TG含量降低。不同孵育时间培育(3、12、24、48 h)后,TG含量随培育时间延长而逐步降低。结论:雪菊黄酮能降低脂肪变性肝细胞中TG的含量,作用效果与雪菊黄酮的作用浓度和培育时间相关,未见明显肝细胞酶学损害。
Objective: To study the lipid-lowering effect of flavonoids from Coreopsis tinctoria on the livercells with fatty degeneration. Methods: The medical fat emulsion injection was used as fat resource to make humanliver fat cell model on normal liver cell line L-02. With different concentrations (20, 50, and 100 μmol/Lflavonoids from Coreopsis tinctoria were co-cultured with hepatocytes with fatty degeneration and incubated fordifferent times (3, 12, 24, 48 h). The oil red staining was used to mark the intracellular lipid droplets and morphologicalchanges. The cell culture supernatants were collected to test triglyceride content. Results: L-02 high fatcell model was feasible. High concentration of flavonoids (100 μmol/L) showed the significant decrease of lipiddroplets and lower triglyceride levels. The triglyceride decreased with prolonged incubation time. Conclusion:Flavonoids from Coreopsis tinctoria can reduce triglyceride in the liver cells with fat degeneration. There is nosignificant liver cell enzyme damage. The effect depends on the concentration and incubation time.
