摘要
目的探讨微小RNA(miRNA-296)对人类风湿关节炎(RA)滑膜成纤维细胞MH7A的影响及机制。方法将miRNA-296抑制物转染人人类风湿关节炎滑膜成纤维细胞MH7A细胞株后,采用Transwell小室法检测细胞侵袭能力变化;实时定量荧光聚合酶链反应(Real-timePCR)检测转染miRNA-296抑制物前后Janus激酶2(JAK2)、信号转导子和转录激活子3(STAT3)、瘦素的mRNA表达变化;Westernblot技术分析miRNA-296抑制物对MH7A细胞中JAK2、STA33、瘦素蛋白的表达影响。结果miRNA-296抑制物对MH7A细胞侵袭有明显的抑制作用。下调miRNA-296能够抑制MH7A细胞中JAK2、STAT3mRNA和蛋白的表达、提高瘦素mRNA和蛋白的表达。结论低表达miRNA-296可以显著抑制人类风湿关节炎滑膜成纤维细胞MH7A的侵袭能力,这可能与JAK2/STAT3/瘦素信号通路有关。
Objective To explore the effect of microRNA-296 (miRNA-296) on human rheumatoid synoviocyte MH7A and its mechanism. Methods MiRNA-296 inhibitors were transfected into MH7A ceils. The cell invasion capability was determined by transwell chamber assay. The expression of miRNA-296 was detected by real-time PCR. The expression of JAK2/STAT3/leptin mRNA were examined by real-time PCR and their protein levels were examined by Western blotting. Results MiRNA-296 inhibitors significantly inhibited the invasion of MH7A cells. MiRNA-296 inhibitors could inhibit the mRNA and protein expression of JAK2 and STAT3 and enhanced the mRNA and protein expression of leptin. Conclusion Low expression of miRNA-296 significantly inhibits the invasion ability of MH7A cells by JAK2/STAT3/leptin pathway.
