自制研发乳粉对HepG-2细胞胰岛素抵抗改善作用的研究

摘要 体外建立人肝癌细胞(HepG-2)胰岛素抵抗模型,对实验室自制乳粉与3种市售乳粉对胰岛素抵抗的改善作用进行研究。用不同浓度的胰岛素对HepG-2细胞进行不同时间的诱导,通过MTT法对细胞活性进行评价及葡萄糖氧化酶法对HepG-2细胞葡萄糖消耗量进行测定,确定HepG-2胰岛素抵抗模型的胰岛素诱导浓度及时间;通过对细胞模型培养过程中添加不同浓度的乳粉,评价乳粉对胰岛素抵抗的改善作用。结果表明,HepG-2细胞在10-7 mol/L浓度的胰岛素中作用24h,葡萄糖消耗量显著降低(P<0.01),说明模型构建成功;在此胰岛素抵抗模型中添加乳粉样品,得出自制乳粉能够改善胰岛素抵抗现象,说明实验室自制乳粉具有一定体外降糖的作用,可以增强细胞对胰岛素的敏感性。 To establish human hepatoma carcinoma cell(HepG-2 cell) model of insulin resistance in vitro and study the effect of milk power on insulin resistance of HepG-2 cells. The three kinds of milk powder selected from market, the other one made in our lab were selected as samples. With different concentrations of insulin co-culture with HepG-2 cells at different time, by evaluating cell activity with MTT method and glucose consumption with glucose oxidase method, HepG-2 cell model of insulin resistance was established. By adding different concentrations of milk powder samples during the culturing of model cells, glucose consumption were evaluated. As shown in the results, 10-7 mol/L insulin co-culture with HepG-2 cells for 24 h can significantly reduce cell glucose consumption(P0.01), indicates that the model was successfully established. Meanwhile, when co-culture the cell model with concentrations of milk powder samples, there have good effect of improving insulin resistance, indicating milk power's good function of preventing diabetes.
