摘要
建立了HPLC-ELSD法同时测定大豆中果糖、葡萄糖、蔗糖、棉籽糖和水苏糖的方法。大豆粉碎后,用水超声提取,用Prevail carbohydrare为色谱柱、乙腈-水(v:v=70:30)为流动相分离,蒸发光散射检测器检测。结果表明,果糖、葡萄糖、蔗糖、棉籽糖、水苏糖的线性范围为(10-500)mg/L,平均回收率为98.16%-101.21%。该方法操作简单,稳定性和重现性好,结果准确度高,为大豆中水溶性糖含量的测定提供了一种准确、灵敏、快速的分析方法。
We established an HPLC-ELSD method to determine the content of fructose, glucose, sucrose, raffinose and stachyose in soybean. Using water ultrasonic extraction the free sugar after the soybean crushed. Chromatographic separation was performed on a Prevail carbohydrate ES column with the mobile phase of acetonitrile-water(v:v=70:30) with the evaporative light scattering detector. The results showed the linearity range of fructose, glucose, sucrose, raffinose and stachyose were(10~500)g/L, and the average recoveries were 98.16%~101.21%, respectively. The established method is simple, reliable, reproducibility and accurate. And the study can provided an accurate, rapid, sensitive analysis for determining the sugar content in soybean.
