不同剂量抗疏健骨颗粒对去势骨质疏松大鼠BMD及血清中ALT和TRACP 5b的影响 被引量:1

Effect of different doses of Kangshu Jiangu Granules on bone mineral density and serum ALT and TRACP 5b in rats with ovariectomized osteoporosis

摘要 目的观察不同剂量抗疏健骨颗粒对绝经后骨质疏松症SD大鼠骨密度(BMD)、碱性磷酸酶(ALP)和抗酒石酸酸性磷酸酶(TRACP 5b)变化的影响,探讨其治疗绝经后骨质疏松症的分子生物学依据。方法采用随机区组设计,将60只SD大鼠分为5组,给药至12周后检测并分析股骨近端骨组织骨密度以及血清中ALP和TRACP 5b的OD值。结果模型组与假手术组相比骨密度明显下降(P<0.05);抗疏健骨颗粒3个剂量组和模型组的骨密度值均明显低于假手术组(P<0.05);与模型组相比大、中、小剂量组的骨密度均有所升高(P<0.05)。假手术组和抗疏健骨颗粒各剂量组血清中ALT和TRACP 5b的OD值明显低于模型组(P<0.05);而抗疏健骨颗粒3个剂量组之间比较,中剂量组的骨密度高于小剂量组和大剂量组(P<0.05);中剂量组血清ALT和TRACP 5b的OD值低于小剂量组和大剂量组(P<0.05)。结论抗疏健骨颗粒能够提高绝经后骨质疏松大鼠的骨密度,抑制破骨细胞活性,降低骨吸收,对绝经后骨代谢异常具有积极的防治作用。 Objective To observe the effect of different doses of Kangshu Jiangu Granules on bone mineral density( BMD),alkaline phosphatase( ALT) and tartrate resistant acid phosphatase( TRACP 5b) in SD rats with postmenopausal osteoporosis,and to explore the evidence in molecular biology of Kangshu Jiangu Granules for postmenopausal osteoporosis. Methods By using random grouping designing,60 SD rats were divided into 5 groups,and treated with Kangshu Jiangu Granules for 12 weeks,then BMD of proximal femur,and OD values of serum ALT and TRACP 5b were detected. Results BMD of model group were significantly decreased compared with that of the sham operation group( P〈0. 05). BMD of three-dose groups and model group were significantly lowered than that of the sham operation group( P〈0. 05). Compared with the model group,BMD of large,medium and small dose groups were increased( P〈0. 05). OD values of serum ALT and TRACP 5b of the sham operation group and Kangshu Jiangu Granules groups of different doses were significantly lower than that of the model group. BMD of the middle dose group was significantly higher than that of the low dose group and large dose group( P〈0. 05). OD values of serum ALT and TRACP 5b of the middle dose group were significantly lowered than that of the low dose group and high dose group( P〈0. 05). Conclusion Kangshu Jiangu Granules can improve bone density and inhibit osteoclast activity,reduce bone absorption in rats with postmenopausal osteoporosis,and lead to a positive effect on the treatment of postmenopausal abnormal bone metabolism.
