电针对兔易损粥样斑块内PDGF-BB及其受体PDGF-AB影响的研究 被引量:1

摘要 目的:观察电针对动脉粥样硬化易损斑块内血小板源性生长因子-BB(PDGF-BB)及其受体PDGFR-AB的影响,探讨电针稳定斑块的机制。方法:随机将24只实验兔分为两组:假手组(8只)、实验组(16只)。造模方法采用腹主动脉内皮球囊拉伤配合高脂饮食以及蝰蛇毒和组胺药物触发的方法建立兔腹主动脉不稳定斑块模型,8周末再次将实验组随机分为模型组和电针组,历经12周。采用HE染色观察斑块病理形态学及新生血管的结构情况;采用免疫组学法检测腹主动脉斑块内内皮细胞标志物第八因子相关抗原(FⅧRag)的变化;采用Western blot技术检测PDGF-BB及其受体PDGFR-AB变化。结果:实验结果显示模型组多数以易损斑块为主,斑块内新生血管以单层内皮细胞为主,而电针组干预后斑块相对稳定,新生血管周围的周细胞增多;Western blot检测结果,电针可提高斑块内PDGF-BB、PDGFR-AB蛋白表达水平,与模型组相比差异有显著性。结论:电针具有稳定易损斑块的作用,其机制可能与上调斑块内PDGF-BB、PDGFR-AB表达、募集更多周细胞有关。
