基于液质联用的代谢组学方法研究天冬滋补保健作用的活性成分 被引量:1

Study on the Active Components in Asparagus Cochinchinensis with Nourishing Health Care Function by Metabonomics Based on Liquid Chromatography-Mass Spectrometry

摘要 目的:基于液质联用的代谢组学方法探索天冬的滋补保健作用的活性成分。方法:健康Wistar大鼠(雌雄各半)24只随机分对照组和给药组。给药组以3.0 g獉kg^(-1)獉d^(-1)灌胃天冬,正常组用蒸馏水进行灌胃,连续42 d后麻醉,腹主动脉取血,分离血清,以超高效液相色谱-质谱联用技术(UPLC-MS)与主成分分析(PCA)、正交偏最小二乘判别分析(OPLS-DA)相结合检测血清,筛选出与滋补保健有关的内源性生物标记物。结果:在PCA与OPLS-DA模型中,正常组与给药组大鼠血清样本均有显著差异,跟正常组比较给药组共发现5个与滋补保健有关的内源性生物标记物,其中正离子发现5,6-trans-Vitamin D3、去氧皮质酮等两种内源性生物标记物,负离子发现亚油酸、二十二碳六烯酸、硬脂酸等三种内源性生物标记物,且所有标记物的含量均升高。结论:所发现的内源性生物标记物均为机体的有益活性成分,为天冬当的保健作用提供了理论依据。 ABSTRACT Objective: To explore the active components in Asparagus Cochinchinensis with nourishing health care function by mctabenomics based on UPLC-MS. Methods: Totally 24 Wistar rats ( with the equal numl^r of male and female) were randomly divid- ed into two groups with 12 ones in each. The rats in the administration group were administered Asparagus Cochinchinensis (3.0 g·kg^-1·d^-1) and those in the control group were administered distilled water for 42 consecutive days. On the 43rd day, all the rats were implemented anesthesia. The blood was collected from the abdominal aorta followed by centrifuged. The ultra performance liquid chro-matography coupled with mass spectrometry (UPLC-MS), principle component Analysis (PCA) and orthogonal partial least squares discriminant analysis(OPLS-DA) were used to evaluate the changes of endogenous substances in the rats after the intragastric adminis- tration of Asparagus Cochinchinensis. Results: Significant differences between the control group and the administration group were detec- ted out by the PCA and OPLS-DA model. Five metabolites between the control group and the administration group were identified. 5, 6-Trans-vitamin D3 and deoxycort.icosteroneetc were identified by the positive ion mode, and linoleic acid, docosahexaenoic acid and stearic acid were identified by the negative ion mode as the potential biomarkers. The contents of all potential biomarkers were increased. Conclusion: The biomarkers have many beneficial activities, so the Mongolian medicine Asparagus Cochinchinensis has the health care function. The experiment provides theoretical basis for the promotion of Asparagus Cochinchinensis as a health care product.
