摘要 目的:建立高效液相色谱测定谷维素片含量的方法,并分析比较不同生产厂家生产的谷维素片的主要成分。方法:采用Agela Innoval C_(18)色谱柱(250 mm×4.6 mm,5μm),以乙腈-甲醇-二氯甲烷-冰醋酸(50∶50∶0.3∶0.3)为流动相,流速1.0ml·min^(-1),检测波长326 nm,柱温30℃,进样量20μl。检测国产与日产谷维素片主要成分。结果:谷维素在20.10~100.50μg·ml^(-1)范围内线性关系良好(r=1.000 0),平均回收率为101.2%(RSD=0.4%,n=9)。国产谷维素片主要成分相同,主要成分的含量存在差异,日产谷维素片与国产谷维素片比较,主要成分及其含量均存在较大差异。结论:该方法简便、准确可靠,既能用于谷维素片的含量测定,亦能用于谷维素片主要成分的分析比较。 Objective: To establish an HPLC method for the content determination and comparison of oryzanol tablets from different manufacturers. Methods: The determination was performed on an Agela Innoval C18 column(250 mm×4.6 mm,5μm) using the mobile phase composed of acetonitrile-methanol-dichloromethane-glacial acetic acid (50 : 50 : 0.3 : 0.3 ). The flow rate was 1.0 ml·min^-1, the detection wavelength was 326 nm, the column temperature was 30℃ and the injection volume was 20μl. The contents of oryzanol tablets from home and Japan were determined. Results: Oryzanol was in good linearity within the range of 20.10μg·ml^-1 100.50 μg·m1^-1 (r = 1. 000 0). The mean recovery was 101.2% (RSD =0.4%, n =9). The main components in domestic oryza- nol tablets were the same, while the contents showed significant differences. Comapred with those of oryzanol tablets from Japan, the main components and their contents in the tablets from home had notable differences. Conclusion: The method is simple, accurate and reliable, which can be used to determine the content of oryzanol tablets and compare the main components as well.
