摘要
泛素蛋白酶体途径是真核生物体内最重要的蛋白质降解途径之一,其中,目标蛋白的泛素化过程涉及泛素激活酶、泛素结合酶和泛素蛋白连接酶(E3)。目标蛋白识别中起关键作用的是E3,Kip1泛素-促进复合体(KPC)是一种E3复合体,由KPC1和KPC2组成,KPC1在C端含有一个环指结构域作为催化亚基,是E3不可缺少的组成部分,KPC2含有一个类泛素结构域和两个泛素相关结构域。KPC广泛存在于真核生物中,在神经系统疾病、血液系统疾病、肿瘤疾病等方面起着重要的调控作用。
Ubiquitin-proteasome pathway is one of the most important protein degradation pathways in eukaryotes,among which ubiquitinatioa of target proteins involves in the ubiquitin-activating enzyme, the ubiquitin-eonjugating enzyme and the ubiquitin ligase (E3). The ubiquitin ligase plays a key role in the identification of target proteins, Kip1 ubiquitination-promoting complex(KPC) being a ubiquitin ligase com- plex that consists of KPC1 and KPC2. KPC1 contains a RING finger domain at the C-terminus as the catalytic subunit that is the integral part of the ubiquitin ligase E3, and KPC2 contains a ubiquitin-like domain and two ubiquitin-assoeiated domains. KPC is widespread in the eukaryotes, playing an important role of regula- tion in nervous system diseases, blood diseases and neoplastic diseases.
