制首乌总游离蒽醌对兔颈动脉粥样硬化模型中Caspase-3,8,9 mRNA表达的影响 被引量:4

Influence of total free anthraquinone of Radix Polygoni Multiflori for rabbit carotid artery atherosclerotic model for expression of Caspase-3,8 or 9 mRNA

摘要 目的观察制首乌总游离蒽醌在防治兔颈动脉粥样硬化过程中对Caspase-3,8,9 mRNA表达的影响。方法将36只雄性新西兰大白兔随机分成6组:正常对照组、模型组、血脂康组和制首乌总游离蒽醌高、中、低剂量组。免疫损伤法结合高脂饲料喂养建立AS模型,造模周期为12周。用制首乌总游离蒽醌i.g给药4周后,耳缘静脉取血离心,检测血脂水平;麻醉动物后,取颈动脉,用RT-PCR法检测颈动脉中Caspase-3,8,9 mRNA的表达。结果与模型组比,制首乌总游离蒽醌能降低TG的含量,高剂量组P<0.05,中、低剂量组P<0.01,TC、LDL的含量高、中、低剂量组均有显著降低P<0.01,均具有统计学意义;与模型组比,制首乌总游离蒽醌能降低Caspase-3,8,9 mRNA的表达水平,高剂量组P<0.01,而中、低剂量组P<0.05,均具有统计学意义;病理切片结果显示,制首乌总游离蒽醌能延缓动脉粥样硬化斑块的形成。结论制首乌总游离蒽醌具有抑制细胞凋亡延缓动脉粥样硬化作用,该作用机制可能与其可能下调Caspase-3,8,9mRNA的表达有关。 Objective To observe the influence of free anthraquinone in Fallopia Multiflora in prevention and treatment of rabbit carotid artery atherosclerotic process for expression of Caspase-3, 8 or 9 mRNA. Methods 36 male New Zealand white rabbits were randomly divided into 6 groups: normal group, model group, HypoCol group, Radix Polygoni Muhiflori total free anthraqui- none high, medium, low dose group. Immune damage method combined with high fat feed establish atherosclerosis model, build- ing cycle for 12 weeks. Then with total free anthraquinone of Radix Polygoni Multiflori by intragastric administration for animal. To extract the ear marginal vein and centrifugation after 4 weeks, then detect the serum cholesterol levels. After anesthesia ani- mals, shear the cervical aortic, detect the expression of Caspase-3, 8 or 9 mRNA in cervical aortic by RT - PCR method. Results Compared with model group, Radix Polygoni Muhiflori total free anthraquinone can reduce the levels of TG, the high dose group ( P 〈 0.05 ) , middle and low dose group ( P 〈 0.01 ), the TC and LDL levels of high, medium and low dose group were signifi- cantly lower (P 〈 0.01 ) , haven statistically significant; With the model group than that of Radix Polygoni Multiflori total free an- thraquinone can reduce Caspase- 3, 8 or 9 mRNA expression level, high dose group (P 〈 0.01 ), while middle and low dose group (P 〈 0.05 ), statistically significant; Pathological section results show that the total free anthraquinone of Radix Polygoni Muhiflori can slow down the formation of atherosclerotie plaque. Conclusion The Radix Polygoni Multiflori total free anthraqui- none can restrain apoptosis and slow down atherosclerosis, It's possible to downgrade Caspase - 3, 8 or 9 mRNA expression of the mechanism of action.
