摘要
目的探讨富含脯氨酸的蛋白酪氨酸激酶2(Pyk2)在人类结直肠癌中的作用机制。方法构建有正义Pyk2cDNA和反义Pyk2cDNA的质粒pcDNA4.1/Zeocin稳定转染结直肠癌sw480细胞系,利用流式细胞技术(FCM)检测转染组与对照组细胞在凋亡和细胞周期s期细胞分布的差异。结果转染正义Pyk2cDNA质粒的sw480细胞凋亡水平明显高于对照组,s期细胞数显著减少;转染反义Pyk2质粒的sw480细胞凋亡水平明显低于对照组,s期细胞数显著增多。结论 Pyk2可能是通过提高结直肠癌细胞的凋亡水平,降低其增殖水平而发挥作用。
Objective To explore the mechanism of proline‐rich protein tyrosine kinase‐2 (Pyk2) in human colorectal carcino‐ma .Methods Recombined plasmids pcDNA4 .1/Zeocins constructed with sense and antisense Pyk2 cDNA were stably trans‐fected into sw480 cells ,respectively .The changes in apoptosis and cell cycle were determined by Flow cytometry (FCM) .Re‐sults The apoptosis rate of SW480 cells transfected with Pyk2 cDNA plasmid was significantly higher than the control group , the number of cells distributed in s stage decreased significantly ;while the apoptosis rate of cells transfected with antisense Pyk2 plasmid was significantly lower than that of the control group ,and the number of cells distributed in s stage increased sig‐nificantly .Conclusion Pyk2 could up‐regulate the apoptosis level of sw 480 cells ,while lower its proliferation .
