摘要 目的:探讨4-氨基吡啶对肺癌细胞侵袭转移的影响及其分子机制。方法:采用浓度为0、3、4.7或0、3.125、6.25 mmol/L的4-氨基吡啶分别处理SPCA1和PC9细胞48 h,Transwell实验观察肺癌细胞迁移侵袭能力,蛋白质印迹法检测细胞侵袭转移相关的上皮间质转化标志蛋白N-钙黏蛋白、E-钙黏蛋白以及Snail的表达。结果:Transwell迁移实验和侵袭实验结果显示,4-氨基吡啶组细胞穿膜数明显减少(P<0.05)。蛋白质印迹法结果显示,4-氨基吡啶组细胞E-钙黏蛋白表达上调,N-钙黏蛋白和Snail表达下调。结论:4-氨基吡啶通过调控上皮间质转化蛋白的表达抑制肺癌细胞SPCA1及PC9侵袭转移。 Objective:To explore the effect and molecular mechanism of 4-aminopyridine(4-AP)on mi-gration and invasion of human lung cancer cells.Methods:SPCA1 and PC9 cells were cocultured with 4-AP at various concentrations (0,3,4.7 or 0,3.1 25,6.25 mmol/L)for 48 h,respectively.The ability of cell migration and invasion was detected by Transwell migration and invasion assays.Western blotting was used to examine the expression of cell migration and invasion-associated epithelial-mesenchymal transition proteins.Results:Compared with negative controls,4-AP reduced cell migration and invasion in Transwell migration and invasion assays.Meantime,expression of mesenchymal-related genes including Snail and N-cadherin was decreased concomitant with increased expression of epithelial-marker E-cadherin in cells with 4-AP treatment.Conclusion:4-AP may suppress cell migration and invasion in lung cancer cells SPCA1 and PC9 via modulating the expression of migration and invasion-associated epithelial-mesenchymal transi-tion proteins.
