摘要
目的:观察祛瘀通脉汤对家兔深静脉血栓纤溶酶原激活物抑制剂-1(PAI-1)的影响。方法:通过结扎家兔股静脉制备深静脉血栓模型,分为A组祛瘀通脉汤组、B组血塞通组、C组空白对照组,D组正常对照组,于造模后第2天灌胃给药,每天1次,连续14d。检测造模后第2、7、14天各组PAI-1水平,光镜下观察各组血管阻塞程度。结果:造模后第2天A、B、C 3组PAI-1水平较D组均有升高,差异有统计学意义(P<0.05);造模后第7、14天,A、B组PAI-1均数均较C组降低,差异有统计学意义(P<0.05),A、B两组间差异无统计学意义(P>0.05)。A、B、C 3组均有血栓形成,但A、B组2级以上血栓发生率均明显低于C组。结论:祛瘀通脉汤可显著降低血液中纤溶酶原激活物抑制剂-1水平,抑制静脉血栓形成。
Objective: to observe the effect of Quyu Tongmai decoction on plasminogen activator inhibitor- 1( PAI- 1)in rabbits with deep vein thrombosis. Methods: A deep vein thrombosis( DVT) model was established by femoral vein ligation in rabbits. Rabbits were divided into Quyu Tongmai decoction group( A),Xuesaitong group( B),blank control group( C),and healthy control group( D). Drugs were administered by gavage from the next day after the DVT model was established,on a daily basis for 14 consecutive days. The level of PAI- 1 in each group was measured 2,7 and 14 days after the DVT model was established. The degree of vascular occlusion was observed by light microscopy. Results: At 2 days after the model was established,groups A,B and C exhibited significantly higher PAI- 1 levels than group D( P〈0. 05). At 7 and 14 days after the model was established,the mean PAI- 1 levels of groups A and B were both significantly lower compared with that of group C( P〈0. 05); there was no significant difference between groups A and B( P〉0. 05). Thrombosis was observed in all the three groups,while the incidence rates of grade 〉2 thrombosis in both groups A and B were significantly lower than that in group C. Conclusion:Quyu Tongmai decoction can significantly reduce the serum PAI- 1 level and inhibit vein thrombosis.
