麻桂药浴方熏蒸对腺嘌呤致慢性肾衰竭大鼠模型MMPs活力的影响

Study of MMPs' dynamic change on adenine-induced chronic renal failure rat model by fumigation of Ma Gui decoction

目的探讨麻桂药浴方熏蒸对腺嘌呤致CRF大鼠模型的干预效果,及对MMPs活力的影响。方法将96只SD大鼠随机分为A、B、C、D、E、F组,每组16只。A、B、C、D、E组均予以腺嘌呤致CRF模型,A、B、C组给予麻桂药浴方水煎液40、20、10 ml熏蒸,D组自来水40 ml熏蒸,E、F组不熏蒸,均干预12周。比较干预后血清BUN、Cr、HCT、PLT、GP-ⅡbⅢa水平,行肾脏组织学检查及肾小管评分,行肾组织MMP-2、MMP-9、TIMP-1和TIMP-2 mRNA及蛋白,LN、Ⅳ-C蛋白检测。结果 D、E组血清Cr、BUN及GP-ⅡbⅢa水平,肾小管评分,TIMP-1、TIMP-2蛋白,LN、Ⅳ-C蛋白均高于A、B、C、F组,HCT、PLT水平,MMP-2、MMP-9 mRNA及蛋白均低于A、B、C、F组(P<0.05或P<0.01)。结论麻桂药浴方熏蒸对腺嘌呤致CRF大鼠模型具有较好的保护作用,可能与增加MMPs活力有关。

Objective To explore the intervention effect on adenine-induced chronic renal failure （CRF） rat model by fumigation of Ma Gui decoction, and the influence on matrix metalloproteinases （ MMPs ） vitality. Methods 96 SD rats were randomly divided into group A, B, C, D, E and F, 16 cases per group. Groups A, B, C, D and E were all performed adenine-induced CRF model, groups A, B and C were treated by fumigation with Ma Gui decoction of 40ml, 20ml and 10ml respectively, group D were treated by fumigation with tap water of 40ml, group E and group F were given conventional feeding without fumigation, all groups were treated for 12 weeks. After the intervention, BUN, Cr, HCT, PLT and GP-ⅡbⅢa levels as well as renal histomor- phology and tubular score were compared, MMP-2, MMP-9, TIMP-1 and TIMP-2, laminin （LN） and IV collagen（Ⅳ-C） protein were tested. Results Serum Cr, BUN and GP-ⅡbⅢa level, renal tubular score, proteins of TIMP-1 and TIMP-2, LN and IV-C protein in group D and E were all higher than that in groups A, B, C and F; HCT, PLT, mRNA and proteins of MMP -2 and MMP-9 were all lower than that in groups A, B, C and F （ P〈0.05 or P〈0.01 ）. Conclusion It has good protective effect on adenine-inducod CRF rat model by fumigation of Ma Gui decoction, and it maybe has relation with increasing MMPs vitality.
