摘要
[目的]研究未名玫瑰葡萄秋水仙素处理的最适浓度和时间,及适宜的倍性鉴定方法。[方法]以秋水仙素为诱变剂,利用不同浓度的秋水仙素浸泡种子不同时间,对二倍体葡萄未名玫瑰品种进行加倍诱导,并采用流式分析仪对诱变植株的细胞DNA含量进行鉴定。[结果]从诱导成活率和四倍体诱变率综合考虑,诱变四倍体未名玫瑰种子的适宜条件:当种子胚根长1.0 cm时,用0.5%秋水仙素处理96 h,其成活率和四倍体率分别为11.33%和5.12%。[结论]用倍性分析仪测定细胞倍性是一条简便快捷的鉴定方法。处理时间是影响成活率、四倍体率和变异率的主要因素。
Objective] To research the optimal concentration and time of colchicine treatment for Weiming Meigui Grape,and the optimal ploidy identification method.[Method] With colchicine as the mutagenic agent,seeds were soaked in different concentrations of colchicine for different time,so as to induce the Vitis vinifera.Cell DNA of the induced plants were identified by flow cytometry.[Result] Considering the induction sur-vival rate and tetraploid mutation rate,the optimal condition for tetraploid induction ( Weiming Meigui seeds) was when seed radical was 1 cm in length,it was treated by 0.5% colchicine for 96 h,the rate of tetraploid was 5.12%and the survival rate was 11.33%.[Conclusion]The ploidy analyzer is a simple,rapid method for determining cell ploidy.Treatment time is the main factor affecting the survival rate,tetraploid rate and mu-tation rate.
