血清miRNA-155、miRNA-26a检测在儿童支气管哮喘气道重塑发生中的应用 被引量:5

The detection of miRNA-155 and miRNA-26a for the children with bronchial asthma in airway remodeling in the clinic application

摘要 目的探讨血清微小RNA(miRNA)-155、miRNA-26a检测在儿童支气管哮喘气道重塑发生中的临床应用价值。方法收集58例支气管哮喘患儿的血清及临床资料,将哮喘患儿分为重塑组和非重塑组,其中重塑组30例,非重塑组28例,并选取20例呼吸系统炎症疾病患儿作为对照组,采用实时荧光定量PCR(RT-PCR)技术检测血清miRNA-155、miRNA-26a的表达。比较分析血清miRNA-155、miRNA-26a的变化与哮喘重塑发生之间的关系。结果重塑组血清miRNA-155的表达水平低于非重塑组和对照组,血清miRNA-26a的表达水平高于非重塑组和对照组,差异均有统计学意义(P〈0.01)。重塑组血清miRNA-155、miRNA-26a水平呈负相关(r=-0.652,P〈0.01),且该组两标志物分别与支气管黏膜网状基底膜厚度及成纤维细胞数目有明显相关性(miRNA-155:r=-0.732、0.787,P〈0.01;miRNA-26a:r=-0.690、0.735,P〈0.01)。结论检测血清miRNA-155、miRNA-26a表达可预测儿童支气管哮喘气道重塑的发生,估计病情的发展状况,并为哮喘患儿的早期干预治疗、气道炎症的有效控制及哮喘病程进展的逆转提供依据。 Objective To explore the clinic significance of the detection of miRNA-155 and miR- NA-26a in serum for the children with bronchial asthma in airway remodeling. Methods The serum and clinic data of 58 cases of children with bronchial asthma were collected, divided into remodeling and non-re- modeling groups to use the real-time fluorescent quantitative PCR (Real time-PCR, RT-PCR) technology to detect the expression of miRNA-155 and miRNA-26a in the children's sera. At the same time, the control group was set up with comparative analysis of the changes in the above two markers for the children with a- cute myeloid leukemia. Results The expressions of miRNA-155 and miRNA-26a in the remodeling group compared correspondingly to that in the non-remodeling and control groups, the difference had statistical sig- nificances ( P 〈0. 01 ). The expressions of miRNA-155 and miRNA-26a had a negative correlation in the remodeling group ( r = - 0. 652, P 〈 0. 01 ), which was associated obviously with thickness of bronchial mucosa reticular basement membrane and the number of fibroblasts ( r = - 0. 732,0. 787 ,P 〈 0. 01 ), and ( r = - 0. 690,0. 735, P 〈 0. 01 ). Conclusions The detection of the expressions of miRNA-155 and miRNA-26a can forecast the occurrence of reshaping for children on the disease with bronchial asthma, esti- mated the development of the disease to provide basis for early intervention and treatment for children with asthma in effective controlling of airway inflammation and reversing the progression of asthma.
