摘要
目的:研究IL-24基因与NCTD联合作用对A549细胞增殖和凋亡的影响。方法:IL-24和(或)NCTD干预A549细胞24 h后,MTT法检测A549细胞增殖抑制率;用流式细胞仪检测细胞凋亡及周期的变化;Western blot法检测Bax、Bcl-2、VEGF和HSP-70蛋白表达水平。结果:IL-24联合NCTD作用24 h对A549细胞增殖抑制率为60.3%,与NCTD/IL-24组比较差异有显著性;IL-24联合NCTD作用对A549细胞凋亡率为32.5%;IL-24和(或)NCTD作用A549细胞后Sub-G1期细胞增多;NCTD/IL-24作用24 h后Bax表达上调,Bcl-2、VEGF表达下调,联合组Bax蛋白表达上调,Bcl-2、VEGF、HSP-70表达下调,尤其VEGF、HSP-70表达下调明显。结论 :IL-24联合NCTD对A549细胞有协同抑制作用。
Objective To investigate the combined effect of IL-24 gene and NCTD on A549 cell. Methods After the IL-24 and NCTD intervening A549 cells, cell proliferation was detected by MTF assay and the changes in apoptosis and cell cycle of A549 was detected by flow cytometry. The expression of Bax, Bcl-2,VEGF and HSP-70 in A549 cells were detected by Western blot. Results The treatment of IL-24 plus NCTD of 24 h resulted in a 60.3% growth in hibition on A549, which showed a significant difference to group intervened by NCTD or IL-24 alone. And the apoptosis rates of A549 was 32.5% after treatment of IL-24 plus NCTD. Furthermore, cell cycle detection indicated that the percentage of A549 in Sub-G1 phases increased in group intervened by IL-24 and/or NCTD. NCTD or IL-24 up-regulated the expression of Bax and down-regulated the expression of Bcl-2 and VEGF after 24 h. Bax expression up-regulated in combination group, and the expression of Bcl-2, VEGF and HSP-70, especially VEGF and HSP-70, down-regulated. Conclusion The combination of IL-24 and NCTD has a synergistic effect on the growth of A549 cells.
