木姜子提取物通过促进E-cadherin表达抑制人高转移肺癌95-D细胞的侵袭转移

摘要 目的:观察木姜子乙醇提取物(litsea pungens ethanol extract,LPEE)对人高转移肺癌95-D细胞侵袭转移能力的影响,以及对E-cadherin表达的影响。方法:不同浓度LPEE作用95-D细胞,MTT法测定细胞增殖抑制率;transwell迁移实验和Matrigel侵袭实验检测LPEE对95-D细胞迁移和侵袭能力的影响;PCR和Western Blot检测LPEE作用后E-cadherin基因和蛋白水平的表达变化。结果 :LPEE(≥50μg/m L)可显著抑制95-D细胞穿过transwell小室和Matrigel基质胶的数量(P<0.01);PCR和Western Blot结果表明LPEE(≥50μg/m L)可促进E-cadherin m RNA和蛋白的表达量(P<0.05)。结论:LPEE可通过促进Ecadherin的表达来抑制肝癌细胞的侵袭和迁移能力。 Objective To evaluate the effects of LPEE on migration and invasion of lung cancer 95-D cells, and the influence of the expression of E-cadherin. Method Different concentrations of LPEE were treated on 95-D, and cell proliferation inhibition rate was determined by MTT. The migration and invasive abilityof the 95-D cells were measured by Transwell chamber and Matrigel assays. The mRNA and protein expression of Ecadherin in 95-D cells were determined by PCR and Western blot. Result LPEE (≥50 μg/ mL) significantly suppressed the number of 95-D cells through the transwell chamber and Matrigel matrix (P 〈 0.01 ), and also promoted the expression of E-cadherin compared with DMSO group(P 〈 0.05). Conclusion LPEE can decrease the migration and invasive ability of lung cancer cells via promoting the expression of E-cadherin.
