摘要
[目的]筛选东北扁核木组织培养最适宜的初代、继代和生根培养基。[方法]以东北扁核木的单芽茎段为外植体,采用MS或1/2MS为基本培养基,添加不同浓度的6-BA、IBA、NAA和GA3,探讨不同培养基对东北扁核木芽诱导、增殖和生根效果的影响。[结果]IBA浓度对东北扁核木初代培养的启动率影响显著,6-BA浓度和GA3浓度的影响不显著,因素主次顺序依次为IBA浓度、GA3浓度、6-BA浓度;6-BA浓度对其继代培养的增殖系数影响显著,IBA浓度和NAA浓度的影响不显著,因素主次顺序依次为6-BA浓度、IBA浓度、NAA浓度;IBA浓度对其生根培养的生根指数影响显著,NAA浓度的影响不显著,因素主次顺序依次为IBA浓度、NAA浓度。[结论]东北扁核木组织培养最适宜的初代、继代和生根培养基分别为MS+0.5 mg/L 6-BA+0.2 mg/L IBA、MS+1.0 mg/L 6-BA+0.1 mg/L IBA和1/2 MS+2.0 mg/L IBA+0.1 mg/L NAA。
[Objective] The aim was to screen out the optimal primary culture, subculture and rooting culture medium for Prinsepia sinensis tissue culture.[ Method] With single bud stem segment of Prinsepia sinensis as explant, using MS or 1/2 MS as the basic culture medium, supplemented with different concentrations of 6-BA, IBA, NAA and GA3 , effects of different culture mediums on bud induction, proliferation and rooting were discussed.[ Result] IBA concentration influenced significantly on starting rate of primary culture of Prinsepia sinensis, the or-der of influence degree was IBA concentration〉GA3 concentration〉6-BA concentration;6-BA concentration had obvious influence on prolif-eration coefficient of subculture medium, the order of influence degree was 6-BA concentration〉IBA concentration〉NAA concentration;IBA concentration had a significant effect on rooting index, while NAA concentration had no significant effect.[ Conclusion] The optimal primary culture, subculture and rooting culture mediums for Prinsepia sinensis tissue culture were: MS +0.5 mg/L 6-BA +0.2 mg/L IBA; MS +1.0 mg/L 6-BA+0.1 mg/L IBA;1/2 MS+2.0 mg/L IBA+0.1 mg/L NAA.
