摘要
文中开发了一种利用特异性PCR(聚合酶链式反应)引物组进行分子标记的辅助方法,以实现对汉麻、苎麻和亚麻的韧皮纤维进行分子生物学鉴定。采用收集的汉麻、苎麻、亚麻3种麻类植物的9个亚种提取基因组DNA并进行简化基因组测序,通过建库与筛选得到能用于鉴定3种麻类韧皮纤维种类的3组特异性PCR引物组。结果表明:通过PCR扩增试验验证证明设计的引物组能实现对汉麻、苎麻和亚麻韧皮纤维种类的定性鉴定;使用植物基因组DNA提取试剂盒法可获得高质量的麻类韧皮纤维DNA样品用于后续的分子标记鉴定方法。
A molecular markers assisted method by using a specific PCR(polymerase chain reaction) primer sets was developed to identify hemp, ramie and flax bast fibers. Nine subspecies of hemp, ramie and flax were collected to extract genomic DNA for reduced-representation genome sequencing. By building a database and screening, 3 groups of specific PCR primer sets were obtained. The PCR amplification experiment results demonstrated that the specific primer sets can be successfully identify the bast fibers of hemp, ramie and flax. The results in this research also showed that the kit method can be used to extract the high quality DNA sample for subsequent molecular marker method.
