摘要
目的:观察芍药甘草汤对支气管哮喘SD大鼠Treg/Th17平衡及其相关细胞因子IL-10、IL-17表达的干预作用,探讨芍药甘草汤防治哮喘的作用机制。方法:雄性SD大鼠40只,随机均分为正常对照组、模型组、芍药甘草汤组、地塞米松组、中西医结合组,每组8只。除正常对照组外其余各组大鼠以卵白蛋白致敏并诱发哮喘模型。正常对照组和模型组0.9%氯化钠溶液灌胃,其余组给予对应药物,用Elisa酶联免疫法测定肺匀浆中相关细胞因子IL-10、IL-17的水平测定,用流式细胞仪检测脾脏单个核细胞液中Treg和Th17比例。结果:与模型组比较,各药物组均可显著降低大鼠脾脏单个核细胞液中Th17比例及肺匀浆中IL-17水平(P<0.01),同时能显著升高大鼠脾脏单个核细胞液中Treg比例及IL-10含量(P<0.01,P<0.05);各药物组之间比较Treg比例及IL-10差异无统计学意义;地塞米松组Th17比例及IL-17表达与中西医结合组比较均显著降低(P<0.01,P<0.05),且IL-17显著低于芍药甘草汤组(P<0.05)。结论:芍药甘草汤防治哮喘的作用机制可能与其调节哮喘大鼠Treg/Th17比例失衡作用有关。
Objective: To observe the intervening effects of Shaoyao Gancao Decoction on Treg/Thl7 and the related cytokines as IL-10 and IL-7 in rats with asthma, and to explore its mechanism in treating asthma. Methods: Forty male SD rats were randomly divided into control group, model group, Shaoyao Gancao Decoction group, dexamethasone group and integrated Chinese and western medicine(tcm-wm) group(n=8 for each group). Asthma rat models were induced by receiving intraperitoneal injection of ovalbumin(OVA) and forced inhalation of atomized OVA. Rats in control and model groups received gavage administration with normal saline, and rats in the other group received gavage administration with corresponding drugs. Expression of IL-10 and IL-17 was detected by using Elisa method. The percentage of splenic regulatory T cells(Tregs) and Th17 cells was analyzed by using flow cytometry. Results: Compared with model group, expression of IL-17 in lung homogenate and percentage of splenic Th17 of treatment groups were decreased significantly(P〈0.01), and expressions of IL-10 and percentage of splenic Treg of rats in treatment groups were significantly increased(P〈0.01, P〈0.05). There were no significant differences in expression of IL-10 and the percentage of splenic Treg among the treatment groups. Expression of IL-17 and percentage of splenic Th17 of dexamethasone group were significant lower than that of integrated tcm-wm group(P〈0.01, P〈0.05). Moreover, IL-17 was significantly lower than that of Shaoyao Gancao Decoction group(P〈0.05). Conclusion: Shaoyao Gancao Decoction could regulate the imbalance of Treg/Thl7, which might be one of its mechanisms in preventing and treating asthma.
