摘要
目的:观察清肠愈疡汤对大鼠溃疡性结肠炎(UC)的治疗作用,并探讨其可能机制。方法:将40只鼠龄11-12周,体质量(200±20)g的SD雄性大鼠按体质量随机分为5组,分别为正常组、模型组、清肠组、5-ASA组、香连组。造模并治疗12d后,进行DAI评分,观察结肠病理改变,RT-PCR方法检测结肠组织中肠三叶因子(ITF),核转录因子-κB(NF-κB),血管活性肠肽(VIP)的表达。结果:清肠组:DAI指数在治疗后第4天1.75±0.59、第8天0.83±0.31、第12天为0.59±0.30呈逐渐降低趋势;溃疡愈合程度良好,结直肠病灶、病理形态均有明显改善,均优于其它组;清肠组中大鼠结肠组织中的ITF、VIP、NF-κB m RNA的表达量分别为0.58±0.01、0.69±0.01、3.15±0.03,与模型组、香连组、5-ASA组比较,有显著性差异(P<0.05)。结论:清肠愈疡汤可以显著改善UC大鼠症状、病理形态,通过提高大鼠结肠组织中VIP的含量,抑制NF-κB的持续激活,上调ITF在结肠组织的表达,从而达到促进溃疡愈合的效果。
Objective: To observe the effects of Qingchang Yuyang Decoction on rats of ulcerative colitis(UC) and explore its possible mechanism. Methods: Forty SD male rats aged 11-12 weeks, body weighted(200±20)g were randomly divided into 5 groups: control group, model group, Qingchang group, 5-aminosalicylic acid(5-ASA) group and Xianglian group. After 12 day's modeling and treatment, DAI score, pathological change of colon, expression of intestinal trefoil factor(ITF), nuclear factor-κB(NF-κB), and vasoactive intestinal peptide(VIP) were examined. Results: Qingchang group: DAI index showed gradual decreasing trend on the fourth day(1.75±0.59), eighth day(0.83±0.31), and twelfth day(0.59±0.30) after treatment; The ulcer was healing well. Colorectal lesions and pathological morphology were significantly improved. Therefore, Qingchang group was superior to other groups; it was noted that ITF, VIP, NF-κB m RNA expression in colon tissue of rats in Qingchang group were 0.58±0.01, 0.69±0.01, and 3.15±0.03, compared with the model group, Xianglian group and 5-ASA group, there was significant difference(P〈0.05). Conclusion: Qingchang Yuyang Decoction could significantly improve the symptoms and pathological morphology in UC rats by increasing the content of VIP in the colon tissue of rats, inhibiting NF-κB activation, upregulating ITF expression in colon tissue, and eventually promote the ulcer healing.
