摘要
目的:探讨新加金水六君煎对野百合碱所致大鼠肺心病肺动脉高压肺血流动力学的影响。方法:50只雄性SPF级Wistar大鼠采用腹腔注射野百合碱造大鼠肺心病肺动脉高压模型,14d后随机分为模型组(0.9%氯钠溶液2m L/d灌胃)、硝苯地平组(0.03g/kg)、附子半夏组(2.25g/kg)、金水六君煎组(8.875g/kg)、新加金水六君煎组(8.875g/kg),另设10只未造模大鼠为空白组灌服0.9%氯化钠溶液,连续灌胃15d。分别于造模及治疗前后观察大鼠的一般状态、活动情况及脱毛现象;最后一次给药后24h,麻醉大鼠,测定HR、MAP,RVSP;计算RV/(LV+S)、RV/BW;观察各组大鼠肺组织病理改变并计算WT和WA。结果:模型组肺动脉血管增厚度、炎性浸润、HR显著降低(P<0.01),肺动脉压MAP、右心室收缩压RVSP、右心指数RV/(LV+S)、右室/体质量比RV/BW、肺动脉管壁厚度与血管外径比值WT、肺动脉管壁面积与血管总面积比值WA较正常对照组显著增加(P<0.01),说明模型大鼠肺动脉高压升高;各给药组与模型组相比均能改善肺动脉血管增厚、炎性浸润,降低m PAP、RVSP、RV/(LV+S)、RV/BW、WT、WA,升高HR,肺血管病理观察以新加金水六君煎组效果最好。结论:新加金水六君煎可以改善肺心病肺动脉高压大鼠的肺血流动力学、右心室肥厚程度及肺组织病理改变,具有一定治疗肺心病肺动脉高压的作用。
Objective: To investigate the effects of Xinjia Jinshui Liujun Decoction on rats with pulmonary hypertension induced by monocrotaline. Methods: Fifty male SPF grade Wistar rats were collected and the pulmonary hypertension with pulmonary heart disease rat model was intraperitoneally injected with MCT. The model rats were randomly divided into 5 groups after 14 d, and respectively administrated by gavage with nifedipine(0.03g/kg), Fuzi-Banxia(2.25g/kg), Jinshui Liujun Decoction(8.875g/kg), XJL(8.875g/kg) and the same volume of normal saline, while the control group was set up with 10 healthy normal rats administrated with the same volume of normal saline, once a day for continuous 15d; 24 h after the last administration anesthetized rats, to measure heart rate(HR), mean arterial pressure(MAP), right ventricular systolic pressure(RVSP); right ventricular index [RV/(LV+S)], right ventricle/body weight were calculated, and observe the pathological changes of rat lung tissue and count the WT and WA: Results: In model group, the pulmonary artery thickness, inflammatory infiltration, HR was significantly decreased(P〈0.01), m PAP(RVSP)RV/(LV+S), RV/BW, WT, WA compared to the normal control group was significantly increased(P〈0.01); Each treatment group can improve the pulmonary vascular thickening, inflammatory infiltration,reduce MAP, RVSP, RV/(LV+S), RV/BW, WT, WA and increase HR, especially the Xinjia Jinshui Liujun Decovtion was best. Conclusion: The Xinjia Jinshui Liujun Decovtion probably can improve hemodynamics, right ventricular hypertrophy and pathological change of lung, so which has good therapeutic effect on rats with pulmonary hypertension.
