探索黑蒜提取液在肝癌放射治疗防护作用的机制 被引量:2

Mechanism study of aged black garlic extract on radiological protection in liver cancer treatment

摘要 目的:探讨黑蒜提取液(ABGE)对肝癌放射治疗防护的作用,观察这一机制是否通过抑制炎性复合体的活化而实现。方法:建立人肝癌小鼠移植瘤模型,将30只荷瘤小鼠随机分为空白对照组、照射组、黑蒜+照射组,分别给予0.9%氯化钠溶液、射线照射及黑蒜提取液联合射线照射的处理,5Gy进行照射,照射后7、14、21d处死实验动物,分离出移植瘤。检测小鼠外周血白细胞计数及活化血小板百分比;Western Blot检测炎性复合体Nlrp1和Nlrp3的表达情况;ELISA检测炎性复合体效应因子IL-1β和IL-18在小鼠外周血血清中的浓度变化。结果:照射组的小鼠外周血中白细胞计数及活化血小板百分比于7d后降低,14d最重,随后逐渐减轻。Western Blot检测黑蒜+照射组Nlrp1和Nlrp3蛋白表达量与空白对照组比明显升高,但低于照射组;ELISA结果显示,照射后14d,IL-1β浓度在照射组及黑蒜+照射组中高于同期空白对照组,随后表达量下降。结论:半致死剂量射线照射小鼠后,黑蒜提取液抑制炎性复合体的活化,抑制局部炎性反应,促进血象修复,从而达到放射防护的作用。 Objective: To investigate the radiation protection mechanism of aged black garlic extract(ABGE) in liver cancer treatment and to study whether the mechanism was performed by inhibiting the inflammatory complex. Methods: Firstly, liver tumor mice model was established, 30 tumor-burdened mice were divided into control group, radiotherapy group and EABG combined radiotherapy group, respectively treated with normal saline, radiation and EABG combined radiation, the dosage of radiation was 5Gy 60Co-γ. Then the mice were sacrificed and the transplanted tumors were isolated respectively at 7, 14, 21 d after radiation. The peripheral blood white blood cell count and activated platelet percentage were detected, the expression of inflammatory complex Nlrp1 and Nlrp3 was determined by Western Blot, and the concentration of inflammatory complex IL-1βand IL-18 in peripheral blood serum was determined by ELISA. Results: Peripheral blood white blood cell count and activated platelet percentage in radiotherapy group showed a downward trend since 7d, reached the lowest level at 14 d. Western blot results showed that the Nlrp1 and Nlrp3 expression increased significantly in EABG combined radiotherapy group comparing with the control group, but below radiotherapy group; ELISA results showed that the concentration of IL-1β increased significantly in radiotherapy group [(150.68±17.76) pg/mL] and EABG combined radiotherapy group [(129.52±8.87) pg/mL] at 14 d, then reverted to normal levels. Conclusion: Aged black garlic extract could protect mice from half lethal dose of 60Co-γ with the possible mechanism of inhibiting local inflammatory reaction and promoting hemogram restoration, suggesting the radiological protection of aged black garlic extract in liver cancer treatment.
