蒙医脑-白脉调控针刺对卒中后抑郁模型大鼠行为及前额叶皮质NE、DA、5-HT含量影响 被引量:7

摘要 目的观察蒙医脑-白脉调控针刺对卒中后抑郁(PSD)模型大鼠行为及前额叶皮质单胺递质的含量影响。方法 32只SD雄性大鼠,随机分为假手术组、模型组、蒙医针刺组、盐酸氟西汀组,每组8只。用栓线法建立局灶性脑缺血再灌注大鼠模型,叠加慢性不可预见性刺激与孤养结合造成PSD动物模型。通过糖水偏爱实验和敞箱实验对大鼠行为学评价,采用酶联免疫法检测大鼠前额叶皮质去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)等单胺递质的含量。结果与假手术组比较,模型组大鼠糖水消耗量、敞箱实验中的水平活动次数和垂直运动次数明显下降(P﹤0.05),前额叶皮质NE、DA、5-HT含量明显降低(P﹤0.05)。与模型组比较,蒙医针刺组和盐酸氟西汀组大鼠糖水消耗量、敞箱实验中的水平活动次数和垂直运动次数明显增加(P﹤0.05),前额叶皮质NE、DA、5-HT含量明显增加(P﹤0.05)。蒙医针刺组和盐酸氟西汀组之间比较,无显著差异(P>0.05)。结论蒙医脑-白脉调控针刺改善卒中后抑郁大鼠行为异常,明显提高前额叶皮质NE、DA、5-HT单胺递质的含量,具有抗抑郁作用。 Objective: To observe the effect of Mongolian medicine brain-white pulse regulating acupuncture on post-stroke depression(PSD) model rats behavioral and cortical monoamine neurotransmitters content. Methods: 32 male SD rats were randomly divided into sham operation group,sample group,Mongolian medicine acupuncture group ,fluoxetine hydrochloride group(drug group). There were 8 rats in each group. PSD animal models were buih by the establishrhent of focal cerebral ischemia rats with the bolt line method, under the conditions of chronic unpredictable stress and separation. By sugar water preference test and open box experiment for the evaluation of rats' behaviour, the content of prefrontal cortex norepinephrine (NE), and dopamine (DA), 5-hydroxytryptamine (5-HT) monoamine n were tested by enzyme-linked immunoassay. Results: Compared with the sham group, model group rats sugar water consumption, the number of horizontal movement and vertical movement in the open box experiment decreased obviously (P〈 0.05),the contents of prefrontal cortex NE, DA, 5-HT of rats in model group decreased significantly (P〈 0.05). Compared with the model group, sugar water consumption,the number of horizontal movement and vertical movement in the open box experiment and the contents of prefrontal cortex NE, DA, 5-HT of rats in acupuncture group and drug group increased significantly (P〈 0.05). There was no significant difference (P〉 0.05) between the acupuncture group and the drug group. Conclusion: Antidepressant effect of Mongolian medicine brain-white pulse regulating acupuncture is related to the improvement of behavior in the rat model of depression after stroke and the contents of NE, DA, 5-HT monoamine neurotransmitters in the brain tissue ,which has antidepressant effect.
