摘要
通过多种测试手段对酸肿牛皮中所提取胶原的性质进行了表征,并与脱灰牛皮中所提取的胶原进行了对比。傅里叶红外、紫外光谱及SDS-PAGE凝胶电泳结果显示,酸肿皮提取的胶原与脱灰皮提取胶原的结构基本一致。微量热差示扫描量热结果显示,酸肿皮提取胶原的热稳定性几乎等同于脱灰皮的。不过,Zeta电位测定表明,酸肿皮提取胶原的等电点降低。扫描电镜观察发现其形貌也发生了变化,通透性变差。成纤维测试结果表明,酸肿牛皮提取胶原的成纤维能力有所下降。
Collagen was extracted from acid- swelling bovine hides( Col- ABH),and then the properties were characterized through lots of methods and compared with those of collagen extracted from deliming bovine hides( Col- DBH). Results from Fourier transform infrared spectroscopy,ultraviolet- visible spectroscopy and sodium dodecyl sulphate polyacrylamide gel electrophoresis( SDS- PAGE) indicate that structure of Col- ABH is similar to that of Col- DBH. Ultra- sensitive differential scanning calorimetry results indicate Col- ABH possesses almost the same thermal stability with that of Col- DBH. Nevertheless,Zeta potential curves reveal that the isoelectric point( p I) of Col- ABH is decreased. The morphology of Col- ABH sponge is also changed and its permeability is poorer. Furthermore,the capacity of fibril formation of Col- ABH is inhibited to some extent.
