摘要
为了建立用于定性、定量检测抗球虫药物青蒿散中有效成分青蒿素的超高效液相色谱(UPLC)检测方法,试验采用反相超高效液相色谱法,将供试品用甲醇超声溶解后离心,取上清液,选择ZORBAX SB-C18键合硅胶柱(4.6 mm×250.0 mm,5.0μm),柱温为35℃,流动相为乙腈-水(62∶38),流速为1.0 mL/min,检测波长为210 nm的检测方法。结果表明:青蒿散中青蒿素的含量在0.2~1.2 mg/mL(r=0.999 9)的范围内线性关系良好,平均加样回收率为98.48%,RSD值为0.99%。说明该方法简单、准确、灵敏,精密度、重复性好,可作为青蒿散中青蒿素含量测定参考方法。
To establish an ultra performance liquid chromatography( UPLC) method for qualitatively and quantitatively detecting the active ingredient artemisinin in anticoccidial drug Artemisia annua powder,a reverse- phase ultra high performance liquid chromatography was used in the test. The samples were dissolved in methanol and assisted with ultrasonic,and then the supernatant was fetched after centrifugation. A detecting method was selected using a ZORBAX SB- C18column( 4. 6 mm × 250. 0 mm,5. 0 μm) with colomn temperature of 35 ℃,acetonitrile and water( V ︰ V = 62 ︰ 38) as the mobile phase with the flow rate of 1. 0 m L/min and the detection wavelength of 210 nm. The results showed that the content of artemisinin in Artemisia annua powder showed good linear relationship in the range from 0. 2 to 1. 2 mg/m L( r =0. 999 9). The average recovery was 98. 48%,and the relative standard deviation( RSD) was 0. 99%. The results indicate that the UPLC method is simple,accurate and sensitive,which has good precision and repeatability,and can be used as a reference method for determining the content of artemisinin in Artemisia annua powder.
